Berbamine Exerts Anti-Inflammatory Effects via Inhibition of NF-κB and MAPK Signaling Pathways

Abstract: Background/Aims: This study aimed to investigate the anti-inflammatory activity of Berbamine (BER), a bisbenzylisoquinoline alkaloid extracted from Berberis amurensis (Xiao Bo An), and the underlying mechanisms. Methods: Macrophages and neutrophils were treated with BER in vitro and stimulated with LPS and fMLP. The effects of BER on the expression of pro-inflammatory mediators in macrophages were evaluated with quantitative RT-PCR and ELISA. The effects of BER on the activation and superoxide release of neutr… Show more

“…In particular, tetrandrine was shown to inhibit IκBα and NFκB p65 phosphorylation and this effect account, at least in part, for its anti-inflammatory properties (Gao et al, 2016;Xu et al, 2016). Berbamine, another BBIQ, also exerts anti-inflammatory effects via inhibition of NFκB and MAPK signaling (Jia et al, 2017). Neferine also inhibits the activation of the NFκB signaling pathway and prevents its nuclear translocation (Liu et al, 2018a).…”

“…Posterior inhibition of p38 MAPK/NF‐κB activation is linked with anti‐inflammatory effects (Jia et al, ). Recently, Segat et al () reported that BCAR at 25 mg/kg (p.so.)…”

“…About 500 natural products belong to this large alkaloid class (Weber & Opatz, 2019). Lots of bisbenzylisoquinoline alkaloids, such as Neferine, Tetrandrine, Cepharanthine, Berbamine and Dauricine, have been found to inhibit the activation of macrophages and thus have anti-inflammatory activity (Gao, Feng, Zhang, Wang, & Cui, 2016;Jia et al, 2017;Paudel, Karki, & Kim, 2016;Qiao et al, 2019;Wu, Guo, Min, Pei, & Chen, 2018). However, the structure-activity relationship of these compounds is still unclear.…”
