“…The resulting plasmid, pKF110, was digested with Sacl and Sall, and a 4.9-kb fragment, carrying the truncated relA gene, was cloned into the Sacl-Sall sites of pGP704 to generate pKF111. This plasmid was propagated in E. coli S17-1 and SM10 and transferred to Vibrio recipients by mating E. coli S17-1 with strain S14 (recipient selected by proline prototrophy) and E. coli SM10 with strain S141 (recipient selected by streptomycin resistance) as described previously (46). By selecting for kanamycin-resistant recipients, strains in which the new AreLA::kan allele had recombined into the chromosomes of the Vibrio strains S14 and S141 were found.…”

“…Vibrio strains were grown at room temperature on VNSS agar (27) or LB plates containing 20 g of NaCl per liter (46). The liquid cultures of Vibrio strains were grown in marine minimal medium (MMM) (46) with rapid gyratory shaking in a water bath at 260C. The MMM contained 0.2 or 0.015% (low glucose concentration) glucose, and all constituents of the MMM medium were sterilized by filtration.…”
