Behavioral Characterization ofdmrt3aMutant Zebrafish Reveals Crucial Aspects of Vertebrate Locomotion through Phenotypes Related to Acceleration

Abstract: Vertebrate locomotion is orchestrated by spinal interneurons making up a central pattern generator. Proper coordination of activity, both within and between segments, is required to generate the desired locomotor output. This coordination is altered during acceleration to ensure the correct recruitment of muscles for the chosen speed. The transcription factor Dmrt3 has been proposed to shape the patterned output at different gaits in horses and mice. Here, we characterized dmrt3a mutant zebrafish, which showed… Show more

“…In our analysis, only acceleration of the larval fish was reduced following DINCH exposure. The study by Del Pozzo et al., showed that dmrt3a gene knockout zebrafish larvae has reduced movement and acceleration due to loss of coordination in the spinal interneurons ( Del Pozo et al., 2020 ). This suggests that apart from distance, acceleration is also an important parameter to consider for behavioral or neurological changes.…”

“…Expression of dmrt3a is associated with spinal cord development and fate specification of dorsal interneurons 6 that coordinates locomotion in animals ( Andersson et al., 2012 ). In a recent study, it was demonstrated that dmrt3a -knockout zebrafish has altered behavior with decreased movement, and acceleration ( Del Pozo et al., 2020 ). We observed an increase in acceleration and parallel with this, significantly induced expression of dmrt3a .…”

“…A premature stop codon by DMRT3 mutation in horses changes locomotion patterns, which is favorable for harnessing racing performances ( Andersson et al, 2012 ; Perry et al, 2019 ). Zebrafish studies have also reported that Dmrt3-expressing neurons contribute to locomotor activity ( Del Pozo et al, 2020 ; Satou et al, 2020 ). These results suggest a conserved function of Dmrt3 in the spinal cord of vertebrates.…”

“…All zebrafish larvae were collected, housed, and treated as previously described by del Pozo at el. in [23]. Pigmentation in larvae used for in situ hybridization was inhibited by addition of 0.003% 1-Phenyl-2-thiourea (PTU) to the embryo water.…”
