zOPT: an open source optical projection tomography system and methods for rapid 3D zebrafish imaging

Abstract: Optical projection tomography (OPT) is a 3D imaging alternative to conventional microscopy which allows imaging of millimeter-sized object with isotropic micrometer resolution. The zebrafish is an established model organism and an important tool used in genetic and chemical screening. The size and optical transparency of the embryo and larva makes them well suited for imaging using OPT. Here, we present an open-source implementation of an OPT platform, built around a customized sample stage, 3D-printed parts a… Show more

“…A custom-built Optical Projection Tomography (OPT) system was used for imaging of the zebrafish embryos at 5 dpf that were euthanised with an overdose of MS-222 (300mg/L), fixed and stained with alcian blue [ 35 , 36 ]. The OPT system, reconstruction algorithms, and alignment workflow were based on the previously described method [ 37 ].…”

“…A custom-built Optical Projection Tomography (OPT) system was used for imaging of the zebrafish embryos fixed at 9 dpf and stained cartilage and bone (Sharpe et al, 2002; Zhang et al, 2020). The OPT system, reconstruction algorithms, and alignment workflow were based on the previously described method (Allalou et al, 2017).…”
