Abstract-Batch experiments were carried out to examine the biomass acclimatization during the aerobic biodegradation of 2, 4 dichlorophenol (2, 4 DCP) by Pseudomonas putida immobilized in polyvinyl alcohol (PVA) gel pellets in a bubble column bioreactor. The bacteria was acclimatized to 2, 4 DCP with concentrations of up to 200 mg/l, with and without the addition of glucose. After the acclimatization, experiments were carried out with initial 2, 4 DCP concentrations ranging from 25 to 100 mg/l. In the course of the study, a phenomenon was noticed that can be related to the effect of carbon starvation on the biodegradation ability of the bacterial cells. This paper presents the results on this phenomenon. Starved cells showed higher growth and degradation rates.

Abstract:The present study focuses on the optimization of process parameters of the aerobic biodegradation of 2, 4 Dichlorophenol (2, 4 DCP) by a commercial strain of P. putida immobilized in PVA gel matrix, using design of experiments by Response Surface Methodology (RSM). The degradation rates obtained from these experiments were used to evaluate the effects of the main factors and their interactions during the biodegradation of 2, 4 DCP. An effective quadratic regression model predicting the rate in terms of the main independent variables, namely temperature, initial pH and initial concentration of 2, 4 DCP, was developed. The optimum conditions for DCP degradation were obtained as follows: Temperature 32.6°C, pH 5.0 and initial DCP concentration 70.5 mg L −1 , resulting in a maximum predicted degradation rate of 41.8 mg L. Under these optimized conditions, a degradation rate of 40.1 mg L −1 h −1was experimentally obtained, thus validating the model.

Phenolic compounds are toxic organic pollutants that are released into the environment through wastewater discharged from several industries. Biodegradation is considered as a most suitable ecofriendly approach for the removal of phenolic waste. Experiments were conducted based on response surface methodology in the batch system to evaluate the requirement of optimum inoculum density, phenol concentration, and incubation period for degradation phenol by strain Bacillus cereus PB1 isolated from polluted site. Box–Behnken design used to analyze the interactions among variables for maximum phenol degradation. Results show optimized parameters of 1% (v/v) initial inoculums, 650 mg L−1 of phenol concentration, and 36 hours of incubation period enhance phenol degradation (402 mg L−1). Analysis of variance (ANOVA) was conducted, and the F‐value was 302.77, which implies that the model is significant. The predicted R‐squared of 0.9590 is close to the adjusted R‐squared of 0.9941, and the P value gives the significance of the coefficient and also indicates the pattern of interactions between the coefficients. Further immobilization experiments performed under an optimized condition infer improved phenol degradation by immobilized cells (464.52 mg L−1). An attempt to investigate the supplementation of culture for increasing adsorption efficiency along with thermally treated rice straw proven about 96.56% of phenol removal.
