Essential Oils from Colombian Plants: Antiviral Potential against Dengue Virus Based on Chemical Composition, In Vitro and In Silico Analyses

Currently, there are no therapies to prevent severe dengue disease. Essential oils (EOs) can serve as primary sources for research and the discovery of phytomedicines for alternative therapy. Fourteen EOs samples were obtained by distillation from six plants used in Colombian folk medicine. GC/MS analysis identified 125 terpenes. Cytopathic effect (CPE) reduction assays revealed differences in antiviral activity. EOs of Lippia alba, citral chemotype and carvone-rich fraction; Lippia origanoides, phellandrene chemotype; and Turnera diffusa, exhibited strong antiviral activity (IC50: 29 to 82 µg/mL; SI: 5.5 to 14.3). EOs of Piper aduncum, Ocimum basilicum, and L. origanoides, carvacrol, and thymol chemotypes, exhibited weak antiviral activity (32 to 53% DENV-CPE reduction at 100 µg/mL; SI > 5.0). Cluster and one-way ANOVA analyses suggest that the strong antiviral activity of EOs could be attributed to increased amounts of non-phenolic oxygenated monoterpenes and sesquiterpene hydrocarbons. Docking analyses (AutoDock Vina) predicted binding affinity between the DENV-2 E protein and terpenes: twenty sesquiterpene hydrocarbons (−8.73 to −6.91 kcal/mol), eight oxygenated monoterpenes (−7.52 to −6.98 kcal/mol), and seven monoterpene hydrocarbons (−7.60 to −6.99 kcal/mol). This study reports for the first time differences in the antiviral activity of EOs against DENV, corresponding to their composition of monoterpenes and sesquiterpenes.

Generally, the herpes virus is categorized into HSV-1 and HSV-2, with HSV-1 being transmitted through oral contacts. In contrast, HSV-2 is transmitted during sexual intercourse; hence known as genital herpes. In the infected individual, the majority of HSV infections are asymptomatic, although herpes can cause painful blisters or ulcers. On the other hand, HSV-2 infection increases the possibility of both transmission and contraction of HIV. In order to eradicate these viral infection complications and avoid the possibility of contracting HIV, few drugs have been prescribed for decades when infected with this viral infection. However, the prescribed drugs are not effective in eradicating this virus from infected individuals, which means few virus particles are latent after treatment with these drugs. Therefore, to investigate the novel anti-herpes potential of phytochemicals, the Maestro V13.3 was run with LigPrep, Grid Generation, SiteMap, Glide XP Docking, Pharmachophores and MM-GBSA. Ultimately, the docking result showed that all examined phytocomponents except ellagic acid had good docking values of − 8.321 (epicatechin), − 8.001 (rac 8-prenylnaringenin), − 7.531 (apigenin) and − 7.252 (−D-(+)catechin) exhibited. In this in-silico assessment, we confirmed that the phytochemicals had more potential scores in docking scores, binding affinity, and MM-GBSA scores compared to the corresponding anti-herpes drugs. Apart from the molecular docking and MM-GBSA values, the phytochemicals were found to have good pharmacological potentials through pharmacophore and pharmacokinetic assessments. Moreover, we believe that compounds such as epicatechin, Rac 8-prenylnaringenin, apigenin and -D-(+)-catechin would reveal possible therapeutic effects when tested in-vitro and in-vivo trials. Finally, the present research suggests that although these molecules have such therapeutic potential, a detailed toxicological study of these molecules should be performed in a dose-dependent manner prior to clinical trials.

With a 30-fold increase in incidence over the previous 50 years, dengue fever is now the most widespread viral disease transmitted by mosquitoes in the world. The intricate interaction of the human defense system, hereditary predisposition, and specific bitterness elements is more likely to be the pathogenesis of dengue. There are presently no viable treatments for dengue. Synthetic drugs which are used against this ailment also show major side effects. There must be a deeper understanding of the underlying mechanism generating severe symptoms to develop auguring markers, cutting-edge diagnostics, and treatments and finally a well-rounded and secure antiserum. Hence, the aim is to search for safer and more potent drugs derived from plants. Plants or herbs are mainly targeting replication or its enzyme or specific stereotypes, though an exact mechanism of phytoconstituents interfering with the viral replication is still undiscovered. The present attempt provided the update with the objective to bringing up forward pathophysiological eventualities involved in dengue virus along with the naturally derived treatment relevant to provide the impregnable therapy by evading the noxious symptoms for dengue fever. Governor’s plum, Cryptocarya chartacea, magnolia berry, and Chinese ginger are such plants exhibiting many effective phytoconstituents against DENV and can be further explored for novel drug discovery by medicinal scientists.
