Avian sarcoma leukosis virus receptor-envelope system for simultaneous dissection of multiple neural circuits in mammalian brain

Abstract: Pathway-specific gene delivery is requisite for understanding complex neuronal systems in which neurons that project to different target regions are locally intermingled. However, conventional genetic tools cannot achieve simultaneous, independent gene delivery into multiple target cells with high efficiency and low cross-reactivity. In this study, we systematically screened all receptor-envelope pairs resulting from the combination of four avian sarcoma leukosis virus (ASLV) envelopes (EnvA, EnvB, EnvC, and E… Show more

“…The cloning of ASLV envelopes and their receptors has revealed their specificity (Barnard et al, 2006;Chai and Bates, 2006): TVB S1 for EnvB and EnvE, TVB S3 for EnvB, TVC for EnvC, and TVJ for EnvJ (Smith et al, 1998;Adkins et al, 2000;Elleder et al, 2005;Chai and Bates, 2006). By using these systems, lentiviral vectors pseudotyped with EnvB, EnvC, and EnvE have been reported to infect cells expressing their corresponding receptors (Matsuyama et al, 2015).…”

“…EnvD was excluded from this procedure because the EnvD-packaged virus may infect mammalian cells that lack foreign receptors (Bova et al, 1988). EnvJ was also omitted because TVJ, an EnvJ receptor, is a sodium-proton exchanger that may potentially affect neuronal activity (Chai and Bates, 2006;Matsuyama et al, 2015).…”
