Autophagy is involved in regulating the immune response of dendritic cells to influenza A (H1N1) pdm09 infection

Abstract: SummaryAutophagy can mediate antiviral immunity. However, it remains unknown whether autophagy regulates the immune response of dendritic cells (DCs) to influenza A (H1N1) pdm09 infection. In this study, we found that infection with the H1N1 virus induced DC autophagy in an endocytosis-dependent manner. Compared with autophagy-deficient Beclin-1 +/À mice, we found that bone-marrow-derived DCs from wild-type mice (WT BMDCs) presented a more mature phenotype on H1N1 infection. Wildtype BMDCs secreted higher leve… Show more

“…As ex- In general, degraded protein aggregates and damaged organelles, including mitochondria, are eliminated from cells by autophagy as a quality control mechanism. 34 In general, autophagy blocks the induction of apoptosis, and apoptosis-associated caspase activation shuts off the autophagic process. 27 These proteins selectively recognize autophagic cargo and mediate its engulfment by autophagosomes by binding to LC3 families.…”

“…Interestingly, dendritic cells from autophagy-deficient Beclin-1 (+/-) mice secreted lower levels of proinflammatory cytokines than those from wild-type mice. 34 In general, autophagy blocks the induction of apoptosis, and apoptosis-associated caspase activation shuts off the autophagic process. 35 We showed that pre-treatment with bafilomycin A1 sensitized OECs to PA-induced cell death.…”

“…Autophagy plays important roles in modulating viral replication and antiviral immune response (McFarlane et al, 2011;Mohl et al, 2012;Monastyrska et al, 2013;Paul and Munz, 2016;Radtke et al, 2013). Increasing evidence indicates that autophagy plays both anti-viral and pro-viral roles in the life cycles and pathogenesis of a wide range of viruses (Chiramel et al, 2013;Jackson, 2015;Zang et al, 2016). Previous studies have shown that coronavirus infection is associated with the autophagic process, and that the nonstructural proteins of coronavirus induce the formation of autophagosomes from the endoplasmic reticulum via an omegasome intermediate (Cottam et al, 2011).…”
