Rosa rugosa Thunb. (Rosaceae) is a common ornamental flower distributed in the temperate regions of eastern Asia and widely cultivated in the Yunnan Province 1,2. The petals and buds of R. rugosa are often used as food, incense and as Chinese medicinal materials for the treatment of stomachache, diarrhea and gynecological problems 3. Previous studies have shown the presence of tannins 4 , terpenoids 5-7 and flavonoids 8,9 in this genus. Anti-inflammatory and cytotoxic activities have been observed with selected chemical ingredients isolated from R. rugosa 7,10. Previous studies in our laboratories on this plant have led to the isolation of three new aurones that possess anti-HIV-1 and cytotoxic properties 9. Continuing the effort to search for novel and bioactive metabolites from medicinal plants, we now report the isolation and characterization of a new flavone (1). In this paper, the structure elucidation of 1 and its biological evaluation are described. EXPERIMENTAL Optical rotations were measured with a Horiba SEPA-300 polarimeter. UV spectra were obtained using a Shimadzu UV-2401A spectrophotometer. A Tenor 27 spectrophotometer was used for scanning IR spectra. 1D-and 2D NMR spectroscopic data were recorded on a DRX-500 NMR spectrometer with TMS as internal standard. Chemical shifts (δ) are expressed in ppm with reference to the TMS signal. HRESIMS was performed on a VG Autospec-3000 spectrometer. Semipreparative HPLC was performed on a Shimadzu LC-8A preparative liquid chromatograph with Zorbax PrepHT GF

Clinopodium genus species are popular traditional Chinese medicinal herbs used to treat bruises and swelling and are also purported to improve blood circulation 1,2 . In recent years, several papers have described phytochemistry investigations of various species of clinopodium and it was found to be rich in saponins 3,4 , flavones 5,6 , polyphenols 7,8 , terpenes 9,10 , etc. Clinopodium urticifolium belongs to the Clinopodium genus, which widely distributed in Gansu, shaanxi, ningxia, in northwest China. In previous work, some bioactive compounds were isolated from this plant 8,11,12 . Motivated by a search for bioactive metabolites from this plant, a reinvestigation for the chemical constituents of the whole plant of C. urticifolium were carried out. As a result, a new pterocarpan (1), together with four known pterocarpan derivatives (2-5), were isolated from this plant. In addition, the cytotoxicity of compound 1 was evaluated. Depression, which can be accompanied by many fatal diseases and a low life quality, has become the leading cause of ill health and disability worldwide. However, Chinese Pharmacopoeia, the most authoritative and evidence-based encyclopedia of Traditional Chinese Medicine (TCM), could contain leads and insights into the development of new antidepressant drugs. In this work, nine herbal medicines with ‘dispel melancholy functions’ specifically documented in Chinese Pharmacopoeia have been comprehensively reviewed with respect to clinical trials, and phytochemical and pharmacological aspects. The nine drugs are Rosae Chinensis Flos, Croci Stigma, Albiziae Cortex and Flos, Roase Rugosae Flos, Curcumae Radix, Hyperici Perforati Herba, Cyperi Rhizoma and Bupleuri Radix. The mechanisms of action of their functional antidepressant compounds, including gallic acid, hypericin, kaempferol, crocetin, crocin, quercetin, luteolin, isorhamnetin, curcumin, hyperforin, adhyperforin, catechin, rutin, puerarin, and saikosaponins A and D, have been collected and discussed. These traditional Chinese herbs and their active compounds provide a promising resource to develop effective new antidepressant drugs in future. Moreover, mechanistic investigations, safety verification and large-scale clinical trials are still expected to finally transform such TCM-based antidepressant resources to new drugs for patients suffering from depression.
