Attachment of Con A or extracellular matrix initiates rapid sprouting by cultured leech neurons.

Abstract: The outgrowth of neurites by single identified leech neurons in culture is markedly influenced by the substrate, Extensive sprouting occurs within a few hours on the plant lectin Con A. In contrast, the same neurons grow far more slowly or not at all when plated on vertebrate extracellular matrix proteins, other lectins, or poly(L-lysine). Sprouting on Con A, unlike that on poly(L-lysine), is inhibited by the Con A-specific hapten sugar methyl a-D-mannoside. Another substrate, promoting even more extensive spr… Show more

“…The peripheral motile regions of leech neurites are rich in micro¢laments Retzius and anterior pagoda (AP) neurons were cultured on concanavalin A (ConA), on which they extend broad, veil-like neurites that allow for detailed analysis of the cytoskeleton (Chiquet & Acklin 1986;Neely 1993;Neely & Gesemann 1994). In phase contrast microscopy, phase-dark areas were often situated in the periphery of these lamellipodia and at the base of ¢lopodia (¢gure 1a and ¢gure 2a).…”

“…Briefly, segmental ganglia were removed from the CNS of anaesthetized animals (chlorobutanol 0.15%; Sigma, Milan, Italy). Neuronal cell bodies of the ventral lateral packet were collected and plated onto concanavalin A (Sigma)-coated COC substrates [47]. Cells were maintained in Leibovitz-15 medium supplemented with 0.6% glucose, 2% fetal calf serum and gentamicin 200 mM (Sigma), at 208C for 2 -4 days.…”

“…The number of trials used for averaging was lower than for the records made from cells on Con A. Similar experiments were also made on AP and P cells. In culture they too branch differently on Con A and on leech laminin extract upon which they have finer processes (3 …”

“…One example is the Retzius cell (1), which has a neuromodulatory function in vivo, secretes serotonin, and forms chemical synapses on P mechanosensory cells in culture. On Con A as substrate, Retzius cells sprout neurites with broad flattened growth cones and stout processes that tend to fasciculate, whereas on leech laminin extract as substrate, outgrowing neurites have small growth cones, and their processes are long and extremely fine with multiple branches (3,4).…”
