Atorvastatin and simvastatin as analgesic agents in experimental models

Abstract: Objectives: 1) To evaluate the analgesic effect of atorvastatin and simvastatin. 2) To compare analgesic activity of these with an established drug tramadol. Materials and Methods: Healthy Albino rats were taken. They were randomly divided into 4 groups of six animals each. Group I: Normal Saline (0.5 ml), Group II: Tramadol (10 mg/kg BW), Group III: Atorvastatin (10 mg/kg BW) and Group IV: Simvastatin (10 mg/kg BW). The animals were subjected to 3 different tests at different interval of time. The tests condu… Show more

“…These results are in conjunction with the findings of other studies. [6,7,12] Dwajani et al [7] also found that the analgesic activity of both the statins starts quite early, which was in conjunction to our findings. Hence, they can be used in acute painful conditions.…”

“…All the drugs were administered orally in the following dosage. [7,9] Group I: Received normal saline (NaCl) 0.5 ml -served as control Group II: Received aspirin 300 mg/kg -served as standard Group III:Received atorvastatin 10 mg/kg -served as experimental Group IV: Received simvastatin 10 mg/kg -served as experimental…”

“…Our results contrast with multiple animal studies that used various experimental pain models. For example, dose-dependent anti-nociception has been demonstrated in statin trials on different mouse pain models, such as writhing, tail-flick, 15 oro-facial formalin, and formalin hind paw tests in mice. 14 16-19 Acute and chronic administration of simvastatin, pravastatin, and atorvastatin also exhibited analgesic effects in mice subjected to the hot plate test.…”

“…PPs at a dose of 20 mg/kg per se lacked this activity and did not alter the analgesic action of atorvastatin. Analgesic and anti-inflammatory activities of atorvastatin have been shown previously in different analgesia tests (Garcia et al, 2011;Dwajani et al, 2012;Jaiswal and Sontakke, 2012).…”
