Astrocytic MicroRNAs and Transcription Factors in Alzheimer’s Disease and Therapeutic Interventions

Abstract: Astrocytes are important for maintaining cholesterol metabolism, glutamate uptake, and neurotransmission. Indeed, inflammatory processes and neurodegeneration contribute to the altered morphology, gene expression, and function of astrocytes. Astrocytes, in collaboration with numerous microRNAs, regulate brain cholesterol levels as well as glutamatergic and inflammatory signaling, all of which contribute to general brain homeostasis. Neural electrical activity, synaptic plasticity processes, learning, and memor… Show more

“…This emerging field holds great promise in shedding light on the toxicological mechanisms underlying pollutant exposure. Specifically, a mounting body of evidence suggests that miRNAs play a crucial role in a complex regulatory network, not only in the regulation of downstream target genes but also in their own regulation by upstream genes, transcription factors, and epigenetic inheritance (Nassar et al., 2022; Tumolo et al., 2022; Wang et al., 2022). MiRNA is transcribed by RNA polymerase II to generate pri‐miRNA, a process that is subject to regulation by multiple transcription factors.…”

“…ncRNAs include dicer-dependent microRNAs (miRNA) and short-interfering RNAs (siRNA), shaped by RNA interference pathways [ 42 ]. Literature suggests the presence of microRNAs in astrocytes that regulate glutamatergic and inflammatory signaling, which affect synaptic plasticity [ 43 ]. siRNAs include nucleotides less than thirty in number, while long non-coding RNAs (lncRNA) include more than two hundred nucleotides [ 44 ].…”
