Astaxanthin Sensitizes Low SOD2-Expressing GBM Cell Lines to TRAIL Treatment via Pathway Involving Mitochondrial Membrane Depolarization

Abstract: Carotenoids have been suggested to have either anti- or pro-oxidative effects in several cancer cells, and those effects can trigger an unbalanced reactive oxygen species (ROS) production resulting in an apoptotic response. Our study aimed to evaluate the effect of the well-known carotenoid 3, 3′-dihydroxy-β, β’-carotene-4, 4-dione (astaxanthin, AXT) on glioblastoma multiforme (GBM) cells, especially as a pretreatment of tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL), that was previously… Show more

“…Astaxanthin treatment for prostate cancer cells for 9 days decreases tumor growth by 38% (Ranga Rao et al 2010 ; Sun et al 2020 ). Similar kinds of results were obtained by McCall et al ( 2018 ) for breast cancer in vitro, Shin et al ( 2022 ) in glioblastoma cell lines, and Kim et al ( 2019 ) in colon cancer cells. The polyunsaturated fatty acid derivatives of shrimps have also been studied for their chemopreventive action in both in vitro and in vivo studies.…”

“…At high concentrations (20-40 μM), it promotes apoptosis in different types of human astroglioma cells; however, at low concentrations (4-8 μM), it causes hormesis by promoting the cell cycle progression through CDK activation and increasing proliferation through the suppression of p53 antitumor protein [213]. In a recent report, hormesis at low doses has been revealed to be associated with the antioxidant properties of astaxanthin, and partly by lowering the mitochondrial membrane potential [214].…”

“…Astaxanthin (3,3-dihydroxy-β,β′-carotene-4,4-dione), a carotenoid recognized for its powerful antioxidant capacity, has shown to have anticarcinoma properties in numerous cancer cell types via various signaling pathways. In this way, astaxanthin can enhance the level of proapoptotic proteins like Bax/Bad, reduce antiapoptotic proteins, such as Bcl-2, and can decrease the rate of ROS in glioblastoma multiforme cells [ 117 ]. Additionally, cellulose nanocrystals/nanofibrils loaded with astaxanthin nanoemulsion enhanced the production of ROS in L929 and NIH3T3 cells as in vitro models of skin cancer [ 118 ].…”
