Association of vitamin D receptor gene 3′‐variants with Hashimoto's thyroiditis in the Croatian population

Abstract: Hashimoto's thyroiditis (HT) is the most frequent autoimmune thyroid disease with strong genetic background. Vitamin D receptor (VDR) endocrine system affects immunosuppressive, regulatory and tolerogenic decisions required for induction and maintenance of peripheral immune tolerance. With respect to the biological function of the VDR and functionally plausible gene-expression data, we sought to test whether particular 3'-restriction fragment length polymorphisms (RFLP) and haplotypes previously directly or in… Show more

“…Polymorphisms of the VDR gene were related to HT, as mentioned previously (35). Moreover, polymorphisms of the CYP27B1 hydroxylase gene catalyzing the conversion of 25 hydroxyvitamin D 3 (25(OH)D) to its active form were also associated with HT (69).…”

“…Several genes are known to be associated with the disease occurrence, progression, and severity. Among them are genes coding for human leukocyte antigen (HLA-DR), cytotoxic T-lymphocyte-associated protein 4 (CTLA4), CD40, protein tyrosine phosphatase 22 (PTPN22), interleukin 2 receptor, alpha (IL2RA), and vitamin D receptor (VDR) (17,34,35). The only thyroid-specific gene associated with HT is the gene for Tg (17).…”

“…Horst-Sikorska Vitamin D levels in autoimmune thyroid diseases S Kivity et al 245 et al 47 linked VDR polymorphism to thyroid autoimmunity susceptibility by demonstrating a correlation between F allele carriers of the VDR-FokI polymorphism and Graves' disease. Stefanic et al 48 reported an association between VDR-common haplotypic variants and Hashimoto's thyroiditis. In addition, several studies demonstrated genetic polymorphism of the vitamin D-binding protein among Polish 49 and Japanese 50 patients with AITDs, whereas the CYP27B1 hydroxylase polymorphism was found to predispose to Hashimoto's thyroiditis and Graves' disease in German subjects.…”

“…The significant association of VDR polymorphism with AITD has also indicated a potential role of vitamin D in the pathogenesis of AITD (22)(23)(24). Activated vitamin D modulates autoimmune reactions by regulating T lymphocytes to inhibit both the production and activity of cytokines (5).…”
