Purpose: This study was implemented to investigate the associations between SNP in mature microRNA (miRNA) sequence and lung cancer prognosis and to verify the function of those SNP.Experimental Design: Eight SNPs (rs3746444T>C in hsa-mir-499, rs4919510C>G in hsa-mir-608, rs13299349G>A in hsa-mir-3152, rs12220909G>C in hsa-mir-4293, rs2168518G>A in hsamir-4513, rs8078913T>C in hsa-mir-4520a, rs11237828T>C in hsa-mir-5579, and rs9295535T>C in hsa-mir-5689) were analyzed in a southern Chinese population with 576 patients with lung cancer, and the significant results were validated in two additional cohorts of 346 and 368 patients, respectively. A series of experiments were performed to evaluate the relevancies of those potentially functional SNPs.Results: We found that the microRNA-499 rs3746444T>C polymorphism exhibited a consistently poor prognosis for patients with lung cancer in the discovery set [HR, 1.24; 95% confidence interval (CI), 1.02-1.49; P ¼ 0.028], in the validation set I (HR, 1.31; 95% CI, 1.01-1.71; P ¼ 0.048) and in the validation set II (HR, 1.45; 95% CI, 1.12-1.86; P ¼ 0.004). The adverse effect of CT/CC variants was more remarkable in patients receiving platinum-based chemotherapy. Further functional assays demonstrated that the rs3746444C variant allele influences the expression of several cancer-related genes and affects lung cancer cells' proliferation and tumor growth in vivo and in vitro via the cisplatinum resistance.Conclusion: Our findings suggested that the rs3746444T>C polymorphism in mature miR-499 sequence could contribute to poor prognosis by modulating cancer-related genes' expression and thus involve tumorigenesis and anti-chemotherapy, which may be a useful biomarker to predict lung cancer patients' prognosis.

TREM2 variants and risk of Alzheimer’s disease: a meta-analysis

Recent studies show that heterozygous variant of triggering receptor expressed on myeloid cells 2 (TREM2) increase the risk of Alzheimer's disease (AD) but with inconclusive results. Here, we conducted a meta-analysis to summarize and clarify the association between TREM2 variants and AD, and examined the relationship between TREM2 genetic variant and the etiology of AD. Relevant case-control studies were retrieved and collected according to established inclusion criteria. Odds ratio (OR) and 95% confidence interval (95% CI) were used to estimate the associations between three TREM2 variants (rs75932628, rs104894002, and rs143332484) and AD. In overall meta-analysis, the summary ORs for rs75932628, rs104894002, and rs143332484 were 2.70 [95% CI: 2.24, 3.24; P < 0.001], 7.21 (95% CI: 1.28, 40.78; P = 0.025), and 1.65 (95% CI: 1.24, 2.21; P = 0.001), respectively, indicating that the TREM2 rs75932628, rs104894002, and rs143332484 may contribute to AD risk. However, sensitivity analysis showed that the results of rs104894002 and rs143332484 should be interpreted with caution, and larger sample size, particularly in different ethnicities, are needed to validate the two variants. The current meta-analysis demonstrates that TREM2 is a candidate gene for AD susceptibility, and TREM2 variant rs75932628 may be a risk factor for AD.

Susceptibility of autoimmune diseases in three polymorphisms of infection-associated gene IRAK1

Introduction: Infection of pathogenic microorganisms is an important reason for autoimmune diseases (ADs). Interleukin-1 (IL-1) receptorassociated kinase-1 (IRAK1) is a key mediator in infection immunity, while the gene of IRAK1 is recognized as a risk gene in ADs. Three single nucleotide polymorphisms (SNPs) in IRAK1 (rs3027898, rs1059702, rs1059703) are considered to be associated with ADs risk. However, the results are conflicting. We conducted this study to get more precise estimations. Methodology: PubMed, OvidSP, and CNKI databases (published prior to August 2014) were searched, and data was extracted from eligible studies. The procedure of statistical analysis was performed using STATA 12.0 software. A random effect model or fixed effect model was chosen based on the between-study heterogeneities. Results: Of the studies involved, 11 studies included 10,705 cases (9,865 controls) for rs3027898, 9 studies included 15,005 cases (14,997 controls) for rs1059702, and 7 studies included 8,115 cases (6,815 controls) for rs1059703. Overall, the results showed that there were significant associations with ADs risk in three genetic models for rs3027898 and in four genetic models for rs1059702, but in neither model for rs1059703. Moreover, in stratified analyses, different extents of associations were found in some different genetic models for all three SNPs. Conclusion: Our data demonstrated that these three SNPs (rs3027898, rs1059702, rs1059703) in IRAK1 were associated with ADs risk.
