Association of the Chloroplastic Respiratory and Photosynthetic Electron Transport Chains of Chlamydomonas reinhardii with Photoreduction and the Oxyhydrogen Reaction

Abstract: The hydrogenase-dependent processes, photoreduction and the dark oxyhydrogen reaction, both of which can support CO2 assimilation, were compared with aerobic photosynthesis and respiration for their sensitivity to electron transport inhibitors in cells and intact chloroplasts of Chiamydomomas reiahardii 11-32/6. Photoreduction but not photosynthesis was inhibited in chloroplasts and the oxyhydrogen reaction detected only in cells was inhibited up to 75 and 90%, respectively, by 150 micromolar rotenone, indicat… Show more

“…In this study, fructose was the externally added sugar, because we now report that 14CO2 is evolved from fructose at a rate roughly 3 to 4 times that of glucose. Consistent with this result is the report of Schnarrenberger (28) (22). Rotenone at the highest concentration tested (50 ,uM) inhibited '4C02 evolution in Chlamydomonas and spinach chloroplasts (Fig.…”

“…An uncharacterized NADPH oxidizing system in C reinhardii thylakoids was reported earlier (6) and was interpreted to function in the evolution of H2 from carbohydrates. The oxidation of PQ by 02 has been shown in other studies with Chamydomonas (7,22,26), indicating the components necessary for a respiratory pathway in the algal chloroplast. The 02-dependent evolution of '4CO2 from darkened spinach chloroplasts externally supplied with ["4C]glucose is consistent also with a respiratory pathway in the higher plant organelle (1).…”

“…4 W m-'. One possible explanation for the ineffectiveness of DCMU is the presence of an NADPH-plastoquinone oxidoreductase in the Chlamydomonas thylakoids, which allows the functioning of a PSI-driven cyclic electron transport through NADPH, resulting in a fairly stable NADPH:NADP ratio (Maione and Gibbs, 1986).…”

“…For the oxyhydrogen reaction to occur, a small and tolerable amount of 02 must be introduced into the gas phase. It is known that 1% 02 is suitable for the oxyhydrogen reaction in C. reinhardtii cells (17). If chloroplastic respiratory metabolism is an integral part of the oxyhydrogen reaction, it became essential to determine whether respiration of an externally supplied carbohydrate can function in this microaerobic condition.…”
