BackgroundThe published findings on the link between the resistin (RETN) gene polymorphism and type 2 diabetes mellitus (T2DM) risk are still contradictory. Here, through a meta-analysis, we summarized a more precise evaluation of their connection by synthesizing existing research.MethodsPubMed, Google Scholar, and Web of Science were electronically searched, and all cited sources were manually searched. The heterogeneity of effects was tested and all statistical analyses were performed in Stata 12.0.ResultsA total of 23 studies with 10,651 cases and 14,366 controls on RETN -420C/G polymorphism were included. The overall results showed that the association of RETN -420C/G polymorphism and T2DM susceptibility was not significant [for the allelic model: odds ratio (OR) = 0.98, 95% confidence interval (CI) = 0.87–1.10, pheterogeneity <.001; I2 = 84.6%; for the dominant model: OR = 0.96, 95% CI = 0.80–1.15, pheterogeneity <.001; I2 = 87.1%; and for the recessive model: OR = 0.96, 95% CI = 0.82–1.12, pheterogeneity <.001; I2 = 56.9%] but with high heterogeneity across studies (p <.0001). Meta-regression found that the median age of T2DM participants (using age 50 as the cutoff) could be a factor in the observed variation. The RETN -420C/G polymorphism seems to be linked to an increased risk of T2DM in younger individuals [for dominant: OR = 0.84 (95% CI, 0.72–0.98; pheterogeneity <.001; I2 = 80.9%)] and decreased risk in older people [for dominant: OR = 3.14 (95% CI, 2.35–4.19; pheterogeneity = .98; I2 = 0.0%)].ConclusionsCurrent results found no evidence that the RETN -420C/G variant was linked to T2DM susceptibility, but the patient’s age appears to be a potential factor that contributed to high heterogeneity across studies. Additional high-quality and well-designed investigations are required to confirm these results.

Cardiovascular diseases (CVDs) have been reported to have a complex pathogenesis by a number of studies. Atherosclerosis and inflammation have been established as the main contributors to CVDs. Furthermore, genetic polymorphisms have been identified and found to have a correlation with an individual’s susceptibility to developing CVD. Some of these polymorphisms and corresponding cardiovascular risk (CVR) factors include: C174G (Interleukin (IL)-6 association), methylenetetrahydrofolate reductase (MTHFR) C667T/A1298C (hyperhomocysteinaemia), VII R353Q (coagulation factor VII association) and rs247616/rs1968905/rs1270922 (cholesteryl ester transfer protein (CEPT) - cholesterol metabolism) amongst others. At a time when disease prediction, diagnosis and prognosis are still being investigated, these polymorphisms have the potential for use in these areas as well as opening more opportunities in the understanding of CVD. The objective of this chapter was to review the current knowledge about the relationship between genetic polymorphisms and cardiovascular disease.

Obesity represents a significant risk factor in the development of type 2 diabetes (T2D), a chronic metabolic disorder characterized by elevated blood glucose levels. Significant sex differences have been identified in the prevalence, development, and pathophysiology of obesity and T2D; however, the underlying molecular mechanisms remain unclear. This study aims to identify sex-specific biomarkers in obesity and T2D and enhance our understanding of the underlying mechanisms associated with sex differences by integrating expression data. A systematic review, individual transcriptomic analysis, gene-level meta-analysis, and functional characterization were performed to achieve this aim. Eight studies and 236 subcutaneous adipose tissue samples were analyzed, identifying common and sex-specific biomarkers, many of which were previously associated with obesity or T2D. The obesity meta-analysis yielded nineteen differentially-expressed biomarkers from a sex-specific perspective (e.g., SPATA18, KREMEN1, NPY4R, and PRM3), while a comparison of the expression profiles between sexes in T2D prompted the identification and validation of specific transcriptomic signatures in males (SAMD9, NBPF3, LDHD, and EHD3) and females (RETN, HEY1, PLPP2, and PM20D2). At the functional level, we highlighted the fundamental role of the Wnt pathway in the development of obesity and T2D in females and the roles of more significant mitochondrial damage and free fatty acids in males. Overall, our sex-specific meta-analyses supported the detection of differentially expressed genes in males and females associated with the development of obesity and T2D, emphasizing the relevance of sex-based information in biomedical data and opening new avenues for research.
