Association of FTO rs9939609 polymorphism with serum leptin, insulin, adiponectin, and lipid profile in overweight adults

Abstract: FTO gene polymorphisms are associated with obesity and food intake. This study aimed to investigate the association of FTO rs9939609 polymorphism genotypes with serum glucose, lipid profile and serum hormones level. This cross-sectional study was carried out on 196 randomly selected overweight adults. Anthropometric measurements including weight, height, body mass index (BMI), fat mass, and fat-free mass were assessed. Serum TGs, total cholesterol, HDL cholesterol, LDL cholesterol, glucose and insulin levels w… Show more

“…The effect of FTO gene on biochemicals parameters was described mostly based on SNPs, too [ 55 , 58 , 59 ]. While some studies showed no effect of polymorphisms on the lipid profile [ 58 ], others described that the SNP was significantly associated with low levels of HDL and high levels of LDL, but all other parameters, i.e., TC, TG, and VLDL, were not affected [ 55 ], or significant differences between FTO genotype groups appeared only for serum HDL levels [ 59 ]. In our study, higher expression of FTO gene was associated with a significantly higher concentration of free fatty acids, but the differences in HDL, LDL or triglycerides levels were not significant.…”

“…Mutations in the FTO gene raise blood levels of leptin, a known mediator or growth factor between obesity and colon cancer, which activates a variety of pathways associated with colon cancer ( Drew, 2012 ; Mehrdad et al, 2020a ). In addition, leptin has been suggested as an intermediate link between obesity and breast or prostate cancer ( Stattin et al, 2001 ; Barone et al, 2020 ).…”

“…32 Based on the available literature, we hypothesized that the Aallele of FTO rs9939609 have an association with vitamin D levels in BC patients. However, in the present study, the patients carrying one or two copies of the FTO rs9939609 risk allele had more than 40 ng/dl serum 25 (OH) vitamin D. Vitamin D was reported to be associated with proper insulin secretion and activity, 16 and lower cir- investigation targeting the cause-effect relationship of the risk allele with the molecular signalling pathways such as the oestrogen signalling pathways may help to understand the cause-effect relationship.…”

“…It is possible that part of the positive effects of vitamin D in preventing disease is due to its role in insulin secretion and function. On the other hand, FTO variation was associated with increased reduced peripheral insulin sensitivity 16. An underlying reason behind high serum level of vitamin D in the BC patients with the FTO risk allele can be that theFTO gene polymorphism may counteract the beneficial effects of vitamin D in preventing BC.…”
