Association of ETS1 polymorphism with granulomatosis with polyangiitis and proteinase 3-anti-neutrophil cytoplasmic antibody positive vasculitis in a Japanese population

Abstract: ETS proto-oncogene 1, transcription factor (ETS1) is involved in various immune responses. Genome-wide association studies on systemic lupus erythematosus in Chinese populations identified the association of ETS1 polymorphism in 3' untranslated region, rs1128334A, which was associated with lower ETS1 expression. In view of substantial sharing of susceptibility genes across multiple autoimmune diseases, we examined whether ETS1 is associated with a rare autoimmune rheumatic disease, anti-neutrophil cytoplasmic … Show more

“…Among Han Chinese population, rs3117242 variant of HLA DPB1 allele was more common among GPA patients than controls . In Japanese population, rs1128334A polymorphism in ETS1 proto‐oncogene was found to be significantly higher in GPA patients and PR3 ANCA positive AAV patients . The geographical variations in the prevalence of the disease and the various organ manifestations could be explained based on genetic factors.…”

“…Cytoplasmic-pattern antineutrophil cytoplasmic autoantibodies (C-ANCA) with antigen specificity for proteinase 3 (PR3-ANCA) are sensitive serologic marker for GPA. ETS1 (ETS protooncogene 1) polymorphism was suggested to be associated with GPA and C-ANCA in Japanese population [62]. However, MPO-ANCA can be seen in 10-20% of patients in Europe and greater than 50% in Asia.…”

“…[10][11][12][13] Cited prevalence rates (per million) include 23.7 in Paris; 14 30 in the United States; 15 95.1 in Norway; 12 112 in New Zealand; 16 and 160 in southern Sweden. 17 GPA affects predominantly Caucasians, 10,12 although cohorts in Korea, 18,19 Japan, 20,21 and China 22,23 have been published. The peak incidence is in the fourth through seventh decades of life; [6][7][8]15 children or adolescents are rarely affected.…”
