Association of Aβ with ceramide-enriched astrosomes mediates Aβ neurotoxicity

Abstract: Amyloid-β (Aβ) associates with extracellular vesicles termed exosomes. It is not clear whether and how exosomes modulate Aβ neurotoxicity in Alzheimer's disease (AD). We show here that brain tissue and serum from the transgenic mouse model of familial AD (5xFAD) and serum from AD patients contains ceramide-enriched and astrocyte-derived exosomes (termed astrosomes) that are associated with Aβ. In Neuro-2a cells, primary cultured neurons, and human induced pluripotent stem cell-derived neurons, Aβ-associated as… Show more

“…During in ammation astrocytes are enriched in Cer. They seem to produce the pro-apoptotic Cer d18:1/16:0 (85,86). Further, reactive astrocytes release extracellular vesicle enriched in ceramide that carryAβ peptides (86).These speci c extracellular vesiclesisolated from brains of 5xFAD mice showed to be particularly toxic for neurons.…”

“…The exchange effects induced by such types of intercellular communication can be variable [ 96 ]. In aged AD patients, most exosomes, released by reactive A1 astrocytes, activate various neurotoxic mechanisms, inducing fragmentation and death of neurons [ 96 , 97 ].…”
