Association between the TGFB1 -509C/T and TGFBR2 -875A/G polymorphisms and gastric cancer: a case-control study

Abstract: Abstract. The transforming growth factor-β (TGFβ) pathway plays an important role in various types of human cancer. However, the role of TGFB1 -509C/T and TGFBR2 -875A/G polymorphisms in gastric cancer is controversial. We aimed to investigate the associations between these polymorphisms and gastric cancer susceptibility, clinicopathological parameters and survival. A case-control study was conducted in 1,010 gastric cancer patients and 1,500 healthy controls. Genotypes were determined by PCR-restriction fragme… Show more

“…Our results showed that subjects carrying the genotype TT in the locus − 509 of the TGF-β gene had a decreased risk for GC (Table 2). Our result is consistent with a recent meta-analysis, which also found that TT genotype at the locus − 509 of the TGF-β gene was protective for GC in an Asian population [21], as well as with other studies [22, 23]. In contrast, this TT genotype has been reported as a risk genotype for GC in an Indian and a Chinese population [24, 25].…”

“…Most studies are about the mutations, variants, and polymorphisms of TGFB1 with gastric cancer. For example, the C-509T [ 38 ] and 509C/T [ 39 ] mutations were significantly associated with gastric cancer invasion. Therefore, in this study, by performing bioinformatics analysis, we showed that TGFB1 could also be considered as a diagnostic biomarker for gastric cancer.…”

“…Previously, we found decreased TGF-β1 in serum samples of male CRC patients significantly associated with CC genotype, indicating that -509C/T functional promoter polymorphism (rs1800469) within the TGF-β1 gene ( TGF-Β1 ) increased the cancer risk, particularly for advanced stages[ 14 ]. A noteworthy finding of the study of Xu et al [ 25 ] was that TGF-Β1 -509C/T and TGF-ΒR2 -875A/G gene polymorphisms acted synergistically on the decreased risk of gastric cancer. The risk of gastric cancer was notably lower among subjects who carried both the TGF-Β1 -509C and TGF-ΒR2 -875A allele genotypes.…”
