Association Between Drug Treatments for Patients With Osteoporosis and Overall Mortality Rates

Abstract: IMPORTANCE Previous studies have reported that drug treatments, particularly treatment with bisphosphonates, is associated with reduced overall mortality rates in addition to decreased fracture risk. If so, drug treatments should be recommended for this reason alone, regardless of a patient's risk of fracture.OBJECTIVE To assess whether randomized clinical trials demonstrate that treatment with bisphosphonates, particularly zoledronate, is associated with reduced mortality rates.DATA SOURCES Science Direct, ME… Show more

“…Over‐activated osteoclastogenesis followed by increased levels of inflammatory factors, such as nuclear factor‐κB ligand (RANKL) and tumour necrosis factor‐α (TNF‐α), is the mechanism of osteoporosis development 11‐13 . Mature osteoclasts contain three or more nuclei and are transformed from the monocyte/macrophage lineage upon stimulation by macrophage colony‐stimulating factor (M‐CSF)and RANKL 14 .…”

“…Fourth, information on the use of medication for osteoporosis was not documented in our patients and the possible effects of antiosteoporosis drugs on DThorax and mortality were not investigated. However, a recent meta-analysis reported that osteoporosis treatment was not associated with a reduction in overall mortality (50). Finally, the study population was only Korean.…”

“…The possible cardioprotective effects of bisphosphonates remain under scrutiny, and there is certainly not sufficiently robust, cohesive data to support a recommendation for this class of medications to be used to treat cardiovascular disease, or even to recommend using them to treat osteoporosis in those at high risk of MI or stroke. A recent meta-analysis reported that mortality was not altered by bisphosphonate treatment [87].…”
