Association between Cyclin D1 polymorphism and oral cancer susceptibility: a meta-analysis

Abstract: Data from several case-control studies on the relation between the Cyclin D1 (CCND1) G870A polymorphism and oral cancer susceptibility implicated conflicting conclusions. Thus, a meta-analysis was performed to derive a more precise evaluation of the association. We searched PubMed and Embase for related studies that had been published in English and eight available studies were finally included in the meta-analysis. Odd ratios (ORs) and 95 % confidence intervals (CIs) were calculated for each study. Our meta-a… Show more

“…Although the expression of cyclin D1b has not been studied in oral cancer, some authors (Holley et al , ; Sathyan et al , ) observed expression of transcript b of CCND1 in OSCC, likely indicating that the more oncogenically active form of cyclin D1 (D1b) is also expressed in oral cancer. Wang et al () addressed in a meta‐analysis the relationship between G/A870 polymorphism and oral cancer. No global association was observed with higher oral cancer risk, but G/A870 polymorphism was related to a higher cancer risk in Asian populations.…”

“…Thus, in the population of Taiwan, rs9344 was associated with decreased oral cancer risk [21], while in Brazilian and German populations no association was found [22,23]. Two independent meta-analyses showed an absence of association between rs9344 and oral cancer risk [23,24], while a positive association was noted in an Asian subgroup [24]. Such discrepancies might be due to a limited number of samples, differences in applied criteria for patients, population stratification, as well as ethnic differences in allele and genotype frequencies.…”

“…Different cyclins work together to control the entire cell cycle. Mutations and amplification of the CCND1 gene are observed frequently in many types of tumors and alter the normal cell cycle, possibly contributing to tumorigenesis [54,55]. Moreover, overexpression of cyclin D1 has been found in breast cancer and may serve as a marker for metastasis in clinical treatment [56].…”
