Assessment of GFP Expression and Viability Using the Tali Image-Based Cytometer

Abstract: Single-cell and population information are commonly obtained either by flow cytometry or fluorescence microscopy. However, these two methods provide different information. Flow cytometry gives quantitative multi-parametric information about physical characteristics and staining or expression, but doesn't allow for visualization. Stand-alone fluorescence microscopy provides visual data, but doesn't allow for straightforward quantitative measurements Here, we present a method for performing cell viability and gr… Show more

“…Following overnight incubation, the HCT-116 cells were treated with 2 mL of medium containing STH (0, 3, 6, 9 and 12 mg/mL) and MH (0, 5, 10, 15 and 20 mg/mL) for 24, 48 and 72 h. In the case of LoVo cells, the concentration of STH was 0, 10, 20, 30 and 40 mg/mL and the concentration of MH was 0, 20, 30, 40 and 50 mg/mL. Cell viability was determined by using the Tali™ Viability Kit—Dead Cell Green reagent following the manufacturer’s instructions and as previously reported by Rampele et al [57]. The proportions of viable and dead cells were analyzed by the Tali™ RFP + Viability assay on the Tali™ Image-Based Cytometer.…”

“…The cancer cell growth inhibition test based on MTT-based assessments of mitochondrial activity is mainly used for drug screening and cytotoxicity testing. 26 We first assessed the cytotoxic potential of compounds 5 – 8 by determining their IC 50 values in human cancer cells (Table 1). Metallomacrocycle 7 exhibited good anticancer potentials in AGS, HCT-15, and HepG2 cells.…”
