ABSTRACT. Genetic diversity within Vicia faba L. is key to the genetic improvement of this important species. In this study, morphological traits and RAPD molecular markers were used to assess the levels of polymorphism across 12 Tunisian populations, three major and nine minor from different locations. Analysis of morphological traits indicated that the three major populations showed significant differences and the nine minor populations exhibited considerable variation for most traits. The grain yield of the Alia population could be increased by inoculation. Of the seven primers tested, it was clear that the Cs12 primer would be recommend for genetic diversity analysis of V. faba. Within population genetic diversity exhibited 94% of total diversity. Intra-population genetic diversity (H S ) was 0.16, which was clearly higher than between population genetic diversity (D ST = 0.06) UPG-MA showed a high level of genetic variation between major and minor populations of V. faba L. Particularly the minor populations showed a high level of diversity and was divided into two subclusters. Ltaifia was separated from the other populations. In addition to a high grain yield, I.Z. Backouchi et al. 7588©FUNPEC-RP www.funpecrp.com.br Genetics and Molecular Research 14 (3): 7587-7596 (2015) these populations showed the lowest Nei and Shannon indices (H = 0.08 and I = 0.13) justifying their homogeneity. For these reasons, these cultivars can be considered a selected population. However, the Takelsa population showed the highest Nei and Shannon indices (H = 0.13 and I = 0.21), indicating that this population was the most heterogeneous, which is interesting for breeding programs.

Asian swamp eel is a highly commercial fish, primarily for China and other Asian countries. The aim of this paper is to evaluate the genetic diversity of wild and cultured samples of Asian swamp eel Monopterus albus using ISSR markers. A total of 129 individuals belonging to three wild samples, Xiantao (XT), Huanggang (HG), Xinyang (XY) and three cultured samples, Wuhan (WH), Jingzhou (JZ) and Nanjing (NJ) were randomly selected for genetic analysis. Twelve ISSR primers were used for screening the six populations and 110 loci were obtained. The polymorphic loci were estimated to be 54%, 56.3%, 58.2%, 60.6%, 69.5% and 71% in NJ, WH, JZ, XT, HG and XY samples, respectively. Average heterozygosity value varied from 0.1956 to 0.2449. The three wild samples showed higher genetic diversity than the cultured samples (P < 0.05), including polymorphic bands (PPB), observed number of alleles per locus (No), effective number of alleles per locus (Ne), Nei's gene diversity index (H ) and Shannon's information index (I ).

Salix lapponum is a relict species that occurs in peat bogs. The most numerous population in Poland, which is located on Lake Bikcze, was selected as a source of plant material for genetic analysis to check if it could be used to translocate and establish new populations in the future. For this purpose, the genetic variability of five groups of plants, which were located 50 to 130 m away from one another, was assessed using the AFLP method. GenoType software did not indicate clonal individuals in the studied population. Although the groups were located at short distances from one another, the genetic differentiation between them, expressed as Φ PT and G ST coefficients, was very high and the clustering methods adopted showed the presence of genetic structure. The lack of correlation between geographic distance and pair-wise Φ PT indicated that at least in the past the gene flow was fluent, but nowadays seedling recruitment is barely visible. These results can be explained by the fact that from the beginning of the 21st century the Lake Bikcze population has dramatically decreased in size. On the other hand, the expected heterozygosity in each group is quite high. If we want to use this population as a donor of reproductive material, action should be taken as quickly as possible.
