Assessment of drug content uniformity of atropine sulfate triturate by liquid chromatography–tandem mass spectrometry, X-ray powder diffraction, and Raman chemical imaging

Abstract: BackgroundAtropine sulfate is an anticholinergic agent for treatment of hypertrophic pyloric stenosis and is orally administrated as a triturate with lactose hydrate. Because of the low safety margin of atropine sulfate, triturate uniformity is a key safety factor. In this study, we assessed the uniformity of atropine sulfate in 1000-fold triturates prepared by wet mixing and dry mixing methods and discussed the cause of the difference in uniformity between two preparation methods.MethodsA 1000-fold triturate … Show more

“…Genetic factors, such as single nucleotide polymorphism Interleukin 7 receptor subunit alpha (IL7RA), may affect predisposition in the development of IRIS [ 29 ]. Allelic polymorphisms (e.g., in CYP2C19 gene) may be considered as patient-specific risk factors affecting fungicidal drug activities, toxicity, and the level of inflammation (e.g., C-reactive protein or albumin levels) [ 30 ]. More recently, transcriptomic profiles have been assessed, and several molecular pathways were proposed (as potential baseline biomarkers), as described below [ 31 ].…”

“…The effect of Raman spectral scattering is weak, and enhancement methods are often used in practical applications, such as surfaceenhanced Raman spectroscopy, high-temperature Raman spectroscopy, resonance Raman spectroscopy, confocal Raman microscopy, Fourier transform Raman spectroscopy, and combinations with other techniques [78]. Modern portable and handheld Raman spectrometers are widely used in drug quality screening to detect low levels of substances or in combination with appropriate stoichiometric methods to distinguish compounds with similar structures and minimal spectral differences [79][80][81][82].…”

“…For this reason, a number of research groups and scientists, including chemists and pharmacists, are working together to establish a valid procedure to estimate a trace amount of ATR, which leads to learning more about ATR, as well as the numerous analytical methods for determining trace amounts, following simple and rapid steps, and understanding its features. As a result, the concept of research was born [1][2][3][4][5].…”
