Aspirin and NSAID use in association with molecular subtypes of prostate cancer defined by TMPRSS2:ERG fusion status

Abstract: Background The TMPRSS2:ERG (T2E) gene fusion is the most common rearrangement in prostate cancer (PCa). It is unknown if these molecular subtypes have a different etiology. We evaluated aspirin and non-aspirin NSAIDs in association with T2E fusion status. Methods Subjects were from a population-based case-control study of PCa. T2E fusion status for prostatectomy cases (n=346) was determined by FISH. Medication use was determined from questionnaires. Logistic regression, controlling for age, race, PCa family … Show more

“…Recently, animal studies and clinical observations (2,3) have found that regular doses of aspirin play a role in the prevention and treatment of colitis-associated colorectal cancer, and could reduce mortality in patients with liver metastasis. In addition, aspirin was found to have an inhibitory effect on the growth of esophageal cancer, ovarian cancer, liver cancer, breast cancer, lung cancer, lymphoma and endometrial cancer (46).…”

“…In a study from our group, we found that the intake of tomato sauce, a substantial contributor to lycopene intake, is more strongly associated with ERG‐positive than ERG‐negative prostate cancer . Similarly, Wright and colleagues determined that the use of aspirin, an anti‐inflammatory medication , is associated with a significantly reduced risk of TMPRSS2:ERG positive prostate cancer only . It could be that tomato sauce and aspirin have properties other than antioxidants that contribute to a reduced risk of TMPRSS2:ERG positive disease.…”

“…Lifestyle may also play a role in the development of PCa defined by TMPRSS2:ERG . For example, lycopene consumption from tomato products has been shown to be inversely associated with ERG‐positive but not ERG‐negative disease, and two studies have shown obesity to be associated with a reduced risk of developing TMPRSS2:ERG ‐positive PCa .…”

“…In the most common known molecular subtype (i.e., roughly half of primary PCa), the oncogene ERG fuses with androgen‐regulated promoter genes, most often TMPRSS2 . Although ERG status is unlikely prognostic by itself, several studies have shown that various risk factors are differentially associated with ERG‐defined PCa . In particular, preliminary evidence suggests that there are distinct inherited genetic factors associated with the risk of ERG‐positive versus ERG‐negative disease, lending plausibility to the hypothesis that family history could be differentially associated with the risk of ERG‐defined disease.…”
