Aptamer-siRNA Chimeras: Discovery, Progress, and Future Prospects

Abstract: Synthetic nucleic acid ligands (aptamers) have emerged as effective delivery tools for many therapeutic oligonucleotide-based drugs, including small interfering RNAs (siRNAs). In this review, we summarize recent progress in the aptamer selection technology that has made possible the identification of cell-specific, cell-internalizing aptamers for the cell-targeted delivery of therapeutic oligonucleotides. In addition, we review the original, proof-of-concept aptamer-siRNA delivery studies and discuss recent ad… Show more

“…There is a disproportionately large number of publications and patents describing aptamer applications compared to the number of publications describing new aptamers [ 79 ]. This trend is consistent across most areas of aptamer research including food safety [ 80 ], neuroscience [ 81 ], medicine [ 82 ] and gene control [ 83 ]. In contrast, the opposite appears to be true in the field of DNA damage and repair; there are fewer examples of aptamer applications compared to the number of selections ( Table 1 ).…”

“…The therapeutic superiority of aptamer-RNA chimeras has been demonstrated in recent years. 56 Wang et al constructed an aptamer-siRNA chimera targeted to epithelial cell adhesion molecule and survivin to deliver a high dose of siRNA and silence resistance to doxorubicin. 57 Silencing of survivin (a key protein involved in drug resistance) resulted in a significant reversal of chemoresistance.…”

“…The idea of a tailored drug for the treatment of a diseased cell avoiding affecting the surrounding healthy cells is not recent, as recalled by Kruspe and Giangrande [54]. It was initially proposed by Paul Ehrlich about one century ago, when he first defined the "magic bullet" and its inherent concept of cell-surface receptor [55].…”
