Aptamers as Versatile Ligands for Biomedical and Pharmaceutical Applications

Abstract: Aptamers are a class of targeting ligands that bind exclusively to biomarkers of interest. Aptamers have been identified as candidates for the construction of various smart systems for therapy, diagnosis, bioimaging, and drug delivery due to their high target affinity and specificity. Aptamers are accounted as chemical antibodies that can be readily linked to drugs, sensors, signal enhancers, or nanocarriers for functionalization. Use of aptamer-guided medications, especially nanomedicines, has resulted in enc… Show more

“…Aptamers are short RNAs or DNAs made up by 20-80 nucleotides. Aptamers can fold into unique three-dimensional conformations to specifically bind to targets (13). F79 synthetic peptide is a member of the aptamers that contains a sequence of 13 amino acids surrounding RAD52 (F79) (VINLANEMFGYNG-GGG-YARAAARQARA), and this was purchased from Genemed Synthesis Inc.. F79 has a three-residue polyglycine linker and protein transduction domain 4; these facilitate passage across lipid bilayers and direct intracellular transduction of the aptamers (14).…”

“…Another class of compound that can be used as a targeting agent are aptamers. They are defined as short RNA or single-stranded DNA oligonucleotides or oligopeptides (5–80 amino acids), which can bind to specific targets through folding into unique three-dimensional conformations [ 143 , 144 ]. Aptamers have unique characteristics such as small size, high stability, structure flexibility, ease of synthesis, and low or no immunogenicity, and these make them a great targeting agent [ 145 ].…”

“…This is the case of the drug pegaptanib, the first aptamer-based drug approved by the FDA, which binds and counteracts the action of VEGF in the treatment of age-related macular regeneration [ 90 , 91 , 92 ]. Importantly, as consequence of their small size and high tissue penetration, aptamers can also be used as delivery cargoes following conjugation with other drugs, such as proteins, small molecules or other oligonucleotides [ 4 , 86 , 93 , 94 , 95 ]. This option, together with high target specificity, represents a huge advantage in the use of oligonucleotide-based therapies, for which delivery problems and off-target effects are still among its main drawbacks precluding further development.…”
