Approaches towards Longevity: Reprogramming, Senolysis, and Improved Mitotic Competence as Anti-Aging Therapies

Abstract: Mainstream approaches that are currently used as anti-aging therapies primarily explore the senescence and epigenetic drift aging hallmarks and they are at two ends of the spectrum. While senolytic therapies include either the selective elimination of senescent cells or the disruption of their secretome with the use of drugs or natural compounds, cellular reprogramming uses genetic manipulation to revert cells all the way back to pluripotency. Here, we describe the progress that has been made on these therapie… Show more

“…Therefore, unapparent phenotypic manifestations of TSM [57], should not be considered as a limitation for defining TSM as a biomarker for multifactorial diseases mediated by X chromosome aneuploidy. Furthermore, aging-related exhausting of molecular pathways guaranteeing chromosome stability and genetic-environmental interactions predispose to an increase in genome instability levels throughout ontogeny [23,34,[58][59][60][61][62]. Moreover, chromosome abnormalities may initiate chromosome instability per se [23].…”

“…As was recently noted, 'the aging genome' is to be protected for successful antiaging therapies [79]. It is more probable that the basis of these therapies is a pathway-based analysis providing an opportunity to define molecular interventions towards longevity in health and disease [80,81]. To perform such an analysis, a closer look at the whole set of genomic changes produced by CIN is required [82,83].…”

“…Senolysis-either the selective elimination of senescent cells or the disruption of their SASP (senescence-associated secretory phenotype) secretome-is one of the key approaches toward longevity and health in old age. 7 Using the bromodeoxyuridine (BrdU) induced model of fibroblast senescence through controlled DNA damage, the inventors identified two macrolide antibiotics of the erythromycin family, azithromycin and roxithromycin, as senolytic drugs. Remarkably, erythromycin showed no such activity whereas telithromycin (a ketolide) did, indicating a complex mechanism that is not dependent on the presence of sugar moieties.…”
