Application of targeted mass spectrometry in bottom-up proteomics for systems biology research

Abstract: The enormous diversity of proteoforms produces tremendous complexity within cellular proteomes, facilitates intricate networks of molecular interactions, and constitutes a formidable analytical challenge for biomedical researchers. Currently, quantitative whole-proteome profiling often relies on non-targeted liquid chromatography-mass spectrometry (LC-MS), which samples proteoforms broadly, but can suffer from lower accuracy, sensitivity, and reproducibility compared with targeted LC-MS. Recent advances in bot… Show more

“…Despite rapid development in MS‐based proteomics over the last 20 years, to date the US Food and Drug Administration (FDA) has approved only around 109 unique protein markers including over 20 protein‐based cancer biomarkers . Proteomics using mass spectrometry has developed into a powerful and highly versatile technology that is enabling rapid advances in diverse areas of biomedical research, clinical diagnostics, and biotechnology . Huh‐7 cells have been used to investigate the liver toxicity of drugs, the molecular mechanisms of hepatic gene regulation, and drug efficacy; hence, we selected Huh‐7 cells as a study model.…”

“…Mass spectrometry with liquid chromatography -mass spectrometry (LC-MS-MS) and matrix-assisted laser desorption/ionization -time of flight (MALDI-TOF/TOF) are extensively applied tools and are principal amongst contemporary protein analysis techniques. 49 1) A systematic review of the literature: In order to gauge the potential of proteomics for biomarker discovery, in February 2020, the PubMed electronic data base was queried by a variety of search terms including, "proteomics and bipolar disorder", "proteomics and mood disorders", "major depressive disorder and proteomic biomarkers", and "schizophrenia and proteomic biomarkers". A large number of articles, totaling more than 500, were recovered.…”
