For ribo¯avin production a highly¯avinogenic strain Eremothecium ashbyii is used. Vitamins are classi®ed as chemical substances that control and effect the physiological processes; Ribo¯avin is one among them. The de®ciency of ribo¯avin in human beings results in the cracking of lips and corners of mouth (cheilosis); nerve tissues are affected. For ribo¯avin production a highlȳ avinogenic strain Eremothecium ashbyii NRRL 1363 was used. Investigations were conducted in shake¯ask using inexpensive and abundantly available raw materials. Among the stimulants, a combination of hog casings and beef extract stimulated the highest and promoted the maximum ribo¯avin yield followed by the combination of ®sh meal and beef extract. The fermented broth (an enriched, ribo¯avin concentrate) can be directly used as a feed grade ribo¯avin. To upgrade it to pharmaceutical grade further investigations are required. SUMMARY Riboflavin [7,8-dimethyl-10-(1′- d -ribityl)isoalloxazine, vitamin B 2 ] is an obligatory component of human and animal diets, as it serves as the precursor of flavin coenzymes, flavin mononucleotide, and flavin adenine dinucleotide, which are involved in oxidative metabolism and other processes. Commercially produced riboflavin is used in agriculture, medicine, and the food industry. Riboflavin synthesis starts from GTP and ribulose-5-phosphate and proceeds through pyrimidine and pteridine intermediates. Flavin nucleotides are synthesized in two consecutive reactions from riboflavin. Some microorganisms and all animal cells are capable of riboflavin uptake, whereas many microorganisms have distinct systems for riboflavin excretion to the medium. Regulation of riboflavin synthesis in bacteria occurs by repression at the transcriptional level by flavin mononucleotide, which binds to nascent noncoding mRNA and blocks further transcription (named the riboswitch). In flavinogenic molds, riboflavin overproduction starts at the stationary phase and is accompanied by derepression of enzymes involved in riboflavin synthesis, sporulation, and mycelial lysis. In flavinogenic yeasts, transcriptional repression of riboflavin synthesis is exerted by iron ions and not by flavins. The putative transcription factor encoded by SEF1 is somehow involved in this regulation. Most commercial riboflavin is currently produced or was produced earlier by microbial synthesis using special selected strains of Bacillus subtilis , Ashbya gossypii , and Candida famata . Whereas earlier RF overproducers were isolated by classical selection, current producers of riboflavin and flavin nucleotides have been developed using modern approaches of metabolic engineering that involve overexpression of structural and regulatory genes of the RF biosynthetic pathway as well as genes involved in the overproduction of the purine precursor of riboflavin, GTP.

A riboflavina ou vitamina B 2 faz parte do complexo B cujo consumo mundial é de aproximadamente 1.250 toneladas por ano. De acordo com a fonte de onde é extraí-da, ela poderá ter várias denominações como: lactoflavina, ovoflavina, hepatoflavina e uroflavina [12]. Suas principais fontes na alimentação são carne, ovo, pescado e leite, apresentando ainda grandes variações de concentração entre as hortaliças e leguminosas. Devese ressaltar ainda a riqueza dessa vitamina nas leveduras. Sua forma livre é um produto natural relativamente raro, ocorrendo em maiores concentrações na retina, urina, leite e fluido seminal de animais, como também em alguns órgãos vegetais [19].Essa vitamina é comercialmente produzida por sín-tese química e por fermentação microbiana, sendo esta última preferida, pois tem propiciado o desenvolvimento de novas tecnologias com diminuição de custos e alta produtividade. Deve ser ressaltado que a riboflavina é a única vitamina que é rápida e totalmente sintetizada em altas concentrações por microrganismos.Com o aumento do consumo de produtos alimentí-cios industrializados, aumenta a preocupação com a perda de nutrientes durante o processo de fabricação dos mesmos, principalmente no que diz respeito às perdas de vitaminas hidrossolúveis, essenciais ao bom funcionamento e desenvolvimento do organismo. A maior parte da riboflavina é usada em formulações farmacêuti-cas, enriquecimento de alimentos e suplementos alimentares para indústrias, de forma a minimizar essas perdas [8,14].Considerando os resultados promissores obtidos utilizando resíduos industriais na composição de meios de cultivo para produção de riboflavina por microrganismo, o aproveitamento de uma matéria bruta de baixo custo (subproduto ou resíduo industrial) é uma alternativa viá-vel na investigação da produção dessa vitamina.O presente trabalho teve como objetivo a produção de riboflavina por processo biotecnológico cultivando Candida guilliermondii DM644 num subproduto de refinaria de óleos vegetais, considerando que há uma carência a nível nacional de produção dessa vitamina e que o mercado consumidor tende a se expandir. Por ou- RESUMOA produção e consumo de alimentos industrializados têm aumentado a preocupação com suplementação e enriquecimento de alimentos com vitaminas e sais minerais, visando repor as possíveis perdas durante os processos de fabricação, principalmente das vitaminas hidrossolúveis, mais especificamente da vitamina B 2 ou riboflavina. Assim sendo, a proposta deste trabalho foi utilizar como componente principal do meio, para produção da riboflavina, um subproduto do refino de óleos vegetais e o microrganismo Candida guillermondii DM 644. A produção da vitamina B 2 foi realizada por fermentação em batelada utilizando Erlenmeyer. As condições empregadas foram agitação orbital, ausência de luz, 30°C, e 24h de incubação.
