Appearance of Segmental Discrepancy of Anion Transport in Rat Distal Colon

Abstract: Anion secretion in mammalian distal colon plays an important role in body electrolyte and water balance under physiological and pathophysiological conditions. Electrogenic Cl Ϫ secretion is the fundamental means of intestinal mucosal surface hydration as a primitive defense mechanism which can prevent the mucosa from noxious stimuli, and several human diseases including secretory diarrhea and cystic fibrosis result from its defective regulation.2) HCO 3 Ϫ , similar to Cl Ϫ , is not only an important component … Show more

“…A study using rat distal colon found that even within this part, segmental differences in anion secretion can be detected. The proximal part of the distal colon displayed a significantly higher HCO 3 Ϫ secretion than the distal part, without changes in mRNA expression of CFTR, NKCC, or NBC, 114) which is in agreement with the lower DRA activity reported in distal colon from rodents. 38) 5.…”

“…The colorectum, a narrow region of the distal colon, is susceptible to pathophysiological stimuli, and also the most vulnerable part to certain colonic diseases. Previous studies have suggested that different colonic segments manifest diverse reactivity to the same agonist or stimulator 19,20 . In the present study, we found that catecholamine (DA and NE)‐induced ion transport was more prominent in the DC1 segment (colorectum) comparing with more proximal regions of the distal colon, e.g., DC4.…”

“…Segmental differences were found even within the distal colon. For example, the segment further away from the anus is more sensitive to 5‐hytroxytryptamine (5‐HT) and acetylcholine than the segment nearest to the anus 19,20 . The colorectum is a narrow region of the most distal colon and is vulnerable colonic diseases and conditions such as stress, inflammation, and ulceration.…”

“…Muscle strips (0.8 cm × 0.2 cm) cut parallel to longitudinal fibers of the colon, each strip with the mucosa and submucosa was carefully removed by scissors and fine forceps, and then suspended in a tissue chamber containing 15 mL Krebs solution. 27 Both ends of each muscle strip were knotted by a medical thread. One end of the strip was fixed to the bottom of the bath with the other connected to a physiological recorder through the tonotransducer.…”
