Antiviral Activity of Oroxylin A against Coxsackievirus B3 Alleviates Virus-Induced Acute Pancreatic Damage in Mice

Abstract: The flavonoids mosloflavone, oroxylin A, and norwogonin, which were purified from Scutellaria baicalensis Georgi, significantly protected Vero cells against Coxsackievirus B3 (CVB3)-induced cell death. To investigate the in vivo antiviral activity of oroxylin A, we intraperitoneally inoculated CVB3 into 4-week-old BALB/c mice. Body weights and blood glucose levels of the mice were decreased after CVB3 infection, and these changes were attenuated by the administration of oroxylin A. Importantly, treatment of mi… Show more

“…Low contents of flavonoids inhibited the replication of Hand Foot Mouth Disease, caused from human enterovirus A71 infection, which can produce severe neurological complications, mainly in young children (Min et al, 2018). Oroxylin A (bacalein-6-methyl ether, an O-methylated flavone), exhibited antiviral activity against Coxsackievirus B3 (Kwon et al, 2016).…”

“…Oroxylin A (5, 7‐dihydroxy‐6‐methoxyflavone, C 16 H 12 O 5 ), the active component of Scutellaria baicalensis , is one of the most important medicinal herbs in traditional Chinese medicine. Oroxylin A has been reported to have antioxidant, anti‐inflammation, anti‐cancer, antiviral and anti‐bacterial infections . Although oroxylin A has been reported to have anti‐hepatic injury effects, the hepatoprotective role of oroxylin A in alcoholic liver injury remains unclear.…”

“…For a 20-μL reverse transcription reaction, RNA was incubated for 60 minutes at 42°C with RNase inhibitor, murine Moloney leukemia virus reverse transcriptase, oligo(dT) 15 primer, and dNTPs in 1× buffer (all from Promega, Madison, WI, USA) in accordance with the manufacturer’s instructions. For the qPCR analysis, the cDNA was amplified by using the CFX96 TM Real-Time PCR Detection System (Bio-Rad, Hercules, CA, USA) with Thunderbird ® SYBR ® qPCR Mix and the following primers: echovirus 30 5′-noncoding region (NCR)-sense, 5′-TCC TCC GGC CCC TGA ATG-3′ and 5′-NCR-antisense, 5′-GAA ACA CGG ACA CCC AAA G-3′ [ 15 ].…”
