Antithrombotic Effect and Mechanism of Radix Paeoniae Rubra

Abstract: The compounds of Radix Paeoniae Rubra (RPR) were isolated and identified by bioassay-guided method, and antithrombotic effects and mechanism were investigated by the acute blood stasis rat model. The RPR extract was evaluated by APTT, TT, PT, and FIB assays in vitro. Results indicated that RPR extract exhibited the anticoagulant activity. In order to find active compounds, six compounds were isolated and identified, and four compounds, paeoniflorin (Pae), pentagalloylglucose (Pen), albiflorin (Ali), and protoc… Show more

“…Further, PGG potently inhibited the enzymatic activity of 1-alkyl-2-acetyl-sn-glycero-3-phosphocholine (PAF) production in rabbit platelet-polymorphonuclear leukocyte (PMN) systems and/or PAF-stimulated platelet activations, 163 and was successful in preventing platelet aggregation (which is an initiating factor for intraluminal thrombus formation in AAA). 117,142 In a similar fashion, PGG extracted from Radix Paeoniae Rubra assisted in downregulation of serum TXB2 levels (thromboxane B2) and upregulated serum 6-Keto-PGF1 α levels (6-keto-pros-taglandin 1 α ) in blood stasis rat models; 193 i.e., it successfully exhibited anticoagulant activity. In addition, PGG improved endothelial NO synthase levels, including hemodynamic metrics such as whole blood viscosity, plasma viscosity, etc .…”

“…From the 10th to 14th day, rats in the control group were injected subcutaneously with 0.9% (w/v) NaCl saline solution, and other rats subcutaneous subcutaneously injected diluted adrenaline hydrochloride (0.67 mg/mL, 0.5 mL/kg) at 10 am, 2 pm, and 6 pm daily. From the 13th to 14th days of the experiment, all rats were soaked in ice-cold water (0~4°C) for 5 min after each injection of adrenaline [ 13 ]. Before the last administration, the rats were fasted for 12 hours but were free to drink water.…”

“…Thrombosis is the formation of thrombus in the vascular system as a result of adhesion of activated platelets and blood coagulation (Xie, Cui, Shan, & Kang, 2017). The triggering of cardiovascular disease by thrombosis because of the activation of hemostasis and hypercoagulability is a major cause of deaths annually (Xie, Zhang, Wang, Wei, & Kang, 2017).…”
