Antisperm Antibodies in Prepubertal Boys and Their Reactivity with Antigenic Determinants on Differentiated Spermatozoa

Abstract: Tests of antibody detection with the use of live sperm (IDIBT and flow cytometry measurements) presented low sensitivity (8% and 48%, respectively) in a group of prepubertal boys. This observation underlines the difficulties in assigning the prospective prognosis of future fertility status in prepubertal boys with antisperm antibodies.

“…In our previous study, we detected ASA in 7% of prepubertal boys (n ¼ 69) with testicular failure (mainly uni-and bilateral cryptorchidism) by means of IDIBT [2]. In the same study, we also detected ASA in some healthy prepubertal boys.…”

“…For each class of immunoglobulin, at least 200 sperm cells were counted and the percentage of positive ones was calculated. A result was considered as positive when at least 10% of sperm cells were attached to the beads [2] at a lower cut-off. Topography of immunobead binding to the particular region of a sperm cell (head, midpiece, tail) was also observed.…”

“…More than 70% of men develop ASAs after vasectomy (17), and there is a limited success in the regain of fertility, even after successful surgical reanastomosis in vasovasostomy attributed to the presence of ASAs (18). Up to 2% to 30% cases of infertility may be associated with the presence of ASAs in the male or female partner of an infertile couple (19,20). These findings provided evidence that spermatozoa can generate an immune response in both men and women, which can lead to a contraceptive state.…”

“…The procedure has been previously described [10]. Briefly, motile sperm cells were isolated from semen of AsA-negative donors by swim-up technique.…”
