Antiproliferative Compounds from Cleistanthus boivinianus from the Madagascar Dry Forest1

Abstract: The two new lignans 3α-O-(β-d-glucopyranosyl)desoxypodophyllotoxin (1) and 4-O-(β-d-glucopyranosyl)dehydropodophyllotoxin (2) were isolated from Cleistanthus boivinianus, together with the known lignans deoxypicropodophyllotoxin (3), (±)-β-apopicropodophyllin (4), (−)-desoxypodophyllotoxin (5), (−)-yatein (6), and β-peltatin-5-O-β-d-glucopyranoside (7). The structures of all compounds were characterized by spectroscopic techniques. Compounds 1, 4, and 5 showed potent antiproliferative activities against the A2… Show more

“…The coupling constant of 3.0 Hz between H-7 and H-8 suggested the cis-geometry of C-7 and C-8 . It was further confirmed by the NOESY interaction of H-7 and H-8 [16].…”

“…As described before in several studies, the phenolic components (flavonoids, flavonoids, phenolic acid, tannins, etc.) revealed a wide range of biological properties such as antibacterial, antiviral, anticancer, anti-proliferative and anti-inflammatory [41][42][43][44][45]. These compounds or secondary metabolites particularly possess also anti-protozoan activities, especially against Plasmodium, Leishmania and Trypanosoma spp.…”

“…The results of DPPH, ABTS +• and FRAP tests of the optimum and the standard compounds (quercetin, oleuropein) were revealed by the antioxidant capacities. However, the difference of quercetin, oleuropein and the optimum of OP extract values could be attributed to a polar character which can contribute to the potentialisation of power to reduce the free radicals [44,65]. According to Miguel [66], some flavonoids having multiple hydroxyl (OH) groups act as pro-oxidants.…”

“…12) 3.2. Diterpenoids Three novel diterpenoids, gravilin (38), dihomoditerpenoid ketone (39), and isodecaryiol (40), along with three known compounds, (+)-(S)-geranyllinalool, (−)-(R)-nephthenol, and 11,12-epoxysarcophytol A, were isolated from the methanol extract of the Madagascan soft coral Sinularia gravis. Compound 38 is a spatane-type diterpenoid, while 39 and 40 are dihomo-and scalarane-type diterpenoids, respectively.…”

“…37) Two new lignans, 3α-O-(β-D-glucopyranosyl) desoxypodophyllotoxin (117) and 4-O-(β-D-glucopyranosyl) dehydropodophyllotoxin (118), were isolated from Cleistanthus boivinianus, together with the known lignans deoxypicropodophyllotoxin, (±)-β-apopicropodophyllin, (−)-desoxypodophyllotoxin, (−)-yatein, and β-peltatin-5-O-β-D-glucopyranoside.Both of the novel compounds exhibited strong antiproliferative activity against the A2780 human ovarian cancer cell line, with IC 50 values of 0.033 and 2.1 µM, respectively. In addition, compound 117 also displayed potent antiproliferative activity against the HCT-116 human colon carcinoma cell line, with an IC 50 value of 0.021 µM 38).…”
