Antioxidative and Whitening Effects of Houttuynia cordata Extract on Lead Acetate of Hair Dye Component

This study was conducted to evaluate the dermal cytotoxicity of lead acetate (LA) and other heavy metal compounds, and the protective effect of Lespedeza bicolor (LB) extract on LA-induced cytotoxicity in cultured B16-/F10 melanoma cells. The study evaluated the antioxidative effects of LB due to its electron-donating ability (EDA), inhibitory effects on melanization and improving cell viability. LA significantly decreased cell viability in a dose-dependent manner, and the XTT50 value was determined at 52.7 μM in the studied cultures. Based on the Borenfreund and Puerner's toxicity criteria, LA was estimated to be highly cytotoxic. LA-induced cytotoxicity and cell damage was reversed by the antioxidant activity of kaempferol (KAE), thereby remarkably improving cell viability. A study of the protective effects of the LB extract on LA-induced cytotoxicity showed that the LB extract remarkably increased cell viability in the LA-treated group, and also inhibited the EDA and the total amount of melanin. The above results suggest oxidative stress-mediated cytotoxicity of LA. In the study, LB extract effectively prevented LA-induced cytotoxicity via its antioxidative activity and inhibition of melanization. In conclusion, natural resources like LB extracts may be useful agents for the prevention of oxidative stress-mediated cytotoxicity and melanization by heavy metallic compounds such as LA.

Protective Effect of Ajuga multiflora BUNGE Extract on Lead Toxicity of Environmental Pollutant

To evaluate the protective effect of Ajuga multiflora BUNGE (AMB) extract on the toxicity of lead acetate (LA), environmental pollutant, cell viability was measured by XTT assay using cultured NIH3T3 fibroblasts. And also, the effect of antioxidant, butylated hydroxytoluene (BHT) on LA-induced cytotoxicity was analysed. For the protective effect of AMB extract on LA-induced cytotoxicity, NIH3T3 fibroblasts were pretreated with 80 or 90 μg/mL of AMB extract for 2 hours before the treatment of LA. And also, the antioxidative effects of AMB extract against LA-induced cytotoxicity were assessed by DPPH-radical scavenging activity, superoxide dismutase (SOD)-like activity and inhibitory activity of lipid peroxidation (LP). In this study, LA significantly decreased cell viability dose-dependently compared with control, and then XTT50 value was determined at 46.1 μM of LA. In the effect of BHT against LA-induced cytotoxicity, it effectively prevented toxic effect of LA by the significant increase of cell viability. In the protective effect of AMB extract on LA-induced cytotoxicity, it significantly increased cell viability which was decreased by LA-induced cytotoxicity, and also it showed the antioxidative effects such as DPPH-radical scavenging activity, SOD-like activity and inhibitory activity of LP. From these results, it is suggested that the cytotoxicity of LA is involved in oxidative stress, and AMB extract effectively prevented the cytotoxicity induced by LA via an antioxidative effect. Conclusively, the natural substance such as AMB extract may be alternative resources for the prevention or treatment of diseases related with oxidative stress.

원광대학교 의과대학 산본병원 내과, 2 원광보건대학교 간호학과This study examined the neurotoxicity induced by lead acetate (LA) on cultured C6 glioma cells and the protective effects of Euphorbiae humifusae L. (EH) extract against LA-induced cytotoxicity. In this study, LA exhibited neurotoxicity in a dose-dependent manner compared to the control, and was determined to be highly-toxic according to the toxic criteria. The XTT50 value of LA was calculated at a concentration of 38.6 M after C6 glioma cells were incubated for 72 hours in the media containing 30∼50 M of LA, respectively. In addition, LA-induced neurotoxicity was suggested to correlate with the level of oxidative stress because vitamin E, an antioxidant, increased the cell viability damaged by LA-induced cytotoxicity. The EH extract effectively prevented cell injury from LA-induced cytotoxicity via its antioxidative effects, such as inhibitory ability of lipid peroxidation, superoxide dismutase-like activity and 1,1-diphenyl-2-picrylhydrazyl-radical scavenging activity. These antioxidative effects may result in components, such as polyphenol or flavonoids including gallic acid or quercetin. In conclusion, natural resources, such as EH extracts, may be a useful putative agent for the treatment of diseases associated with oxidative stress, such as lead neurotoxicity.
