Despite many studies have revealed that developing honey bee (Apis mellifera) larvae are posting a high risk on exposure to insecticides, but the toxicology information on bee larvae remain limited. The present study demonstrated the first assessment of the effects of no observed adverse effect concentration (NOAEC) of carbaryl (CR) and acetamiprid (ACE) on transcriptome and metabolome in honeybee larvae rearing in vitro. Chronic exposure to carbaryl caused a transcriptional response to oxidative stress. In addition, a series of metabolic homeostasis disrupted by carbaryl stress, such amino acid metabolism, purine and pyrimidine metabolism and flavone and flavonol biosynthesis. The activities of enzymic biomarkers including GST, P450, CAT, AChE and SOD were not influenced by ACE stress, while the CR exposure slightly decreased the activity of CAT and SOD. Our results clearly show that ACE and CR varied in their influence on modulation of transcriptome and metabolome, potentially associated with their different toxicity against bee larvae.

At present, a large number of studies have reported that hydrogen has antioxidant functions and prevents oxidative stress damage. However, it is not clear whether hydrogen can prolong longevity based on these effects. Therefore, we studied and explored the antiaging potential of exogenous hydrogen and its ability to extend longevity using Caenorhabditis elegans (C. elegans) as an animal model. Our results showed that the lifespans of the N2, sod-3 and sod-5 mutant strains were extended by approximately 22.7%, 9.5%, and 8.7%, respectively, after hydrogen treatment, but hydrogen had no effect on the lifespans of the daf-2 and daf-16 mutant strains. Meanwhile, the level of reactive oxygen species (ROS) in the hydrogen treatment group was significantly lower than that in the control group. At the transcript level, the expression of age-1 and let-363 was obviously decreased, while the expression of ins-18 was increased at the same time point (14 d). Compared with the control group, paraquat (PQ) could reduce the lifespan of the N2 and sod-5 mutant strains. Importantly, the longevity of these mutant strains recovered to normal levels when the animals were treated with exogenous hydrogen. According to these results, the lifespan of C. elegans is closely related to oxidative stress and can be significantly prolonged by reducing oxidative stress damage. Taken together, our data showed that hydrogen is a valuable antioxidant that can significantly reduce the body's ROS levels and extend the lifespan of C. elegans. This study also laid a foundation for the subsequent application of hydrogen in antiaging studies.

Fluopimomide is an innovative pesticide, widely used for agricultural pest management; however, little is known about its effect on non‐target organisms. This study was designed to assess the potential risk of fluopimomide and the molecular mechanisms using Caenorhabditis elegans, a common model animal. The oxidative stress‐related indicators were analyzed in C. elegans after exposure to fluopimomide for 24 h at three sublethal doses (0.2, 1.0, and 5.0 mg/L). The results demonstrated that sublethal exposure to fluopimomide adversely affected the nematodes growth, locomotive behaviors, reproduction, and lifespan, accompanying with enhanced of reactive oxygen species (ROS) generation, lipid and lipofuscin accumulation, and malondialdehyde content. In addition, exposure to fluopimomide significantly inhibited antioxidant systems including superoxide dismutase, catalase, glutathione S‐transferase, and glutathione in the nematodes. Moreover, the expression of oxidative stress‐related genes of sod‐3, hsp‐16.1, gst‐4, ctl‐2, daf‐16, and daf‐2 were significantly down‐regulated, while the expression of skn‐1 was significantly up‐regulated. Further evidence revealed that daf‐16 and skn‐1 mutant strains of C. elegans significantly decreased ROS production upon fluopimomide exposure compared with the wild‐type nematodes. Overall, our findings indicated that exposure to fluopimomide at sublethal doses caused oxidative damage, mainly associated with insulin/IGF‐1‐like signaling pathway in C. elegans. This is the first report of potential toxic effects of fluopimomide even at low concentrations, providing a new insight into the mechanisms of toxicity to C. elegans by fluopimomide.
