Introduction: The aim of this study was to assess the induction of solvents on the total phenol and flavonoid content and also the antioxidant activity of Ganoderma lucidum extracts. Materials & Methods: In this study, two concentrations (100% and 75%) of diethyl ether, ethanol, butanol, chloroform, and acetone were used as extractants of Ganoderma lucidum. Total phenol and flavonoid contents were measured by spectrophotometric methods and 1, 1-diphenyl-2-picrylhydrazyl (DPPH). Free radical scavenging assay was used for the investigation of antioxidant activity. Results & Discussion: Extractants significantly affected the % yield of extract, the quantity of phenol and flavonoids and antioxidant activity of Ganoderma lucidum mushroom. The highest extraction yield, around 38%, was achieved by 75% acetone, followed by 100% acetone (about 36%) and 75% chloroform (approximate 21%). Hydro-acetone extract exhibited the most significant antioxidative properties (EC50 value; 645.55 µg/mL) comprised of a higher total of phenol content. In conclusion, the total phenol content encouraged the antioxidative potential of Ganoderma lucidum mushroom. Conclusion: These findings indicate that the selective extraction of Ganoderma lucidum shows significant biological activities.

The present paper aims at reviewing and commenting on the analytical methods applied to antioxidant and antioxidant capacity assessment in plant-derived products. Aspects related to oxidative stress, reactive oxidative species' influence on key biomolecules, and antioxidant benefits and modalities of action are discussed. Also, the oxidant-antioxidant balance is critically discussed. The conventional and nonconventional extraction procedures applied prior to analysis are also presented, as the extraction step is of pivotal importance for isolation and concentration of the compound(s) of interest before analysis. Then, the chromatographic, spectrometric, and electrochemical methods for antioxidant and antioxidant capacity determination in plant-derived products are detailed with respect to their principles, characteristics, and specific applications. Peculiarities related to the matrix characteristics and other factors influencing the method's performances are discussed. Health benefits of plants and derived products are described, as indicated in the original source. Finally, critical and conclusive aspects are given when it comes to the choice of a particular extraction procedure and detection method, which should consider the nature of the sample, prevalent antioxidant/antioxidant class, and the mechanism underlying each technique. Advantages and disadvantages are discussed for each method.

Abstract:OBJECTIVE: Despite the great number of substances produced by the skin care industry, very few of them seem to truly have an effect on the skin. Therefore, given the social implications surrounding physical appearance, the search for new bioactive compounds to prevent or attenuate skin aging and enhance self-image is a priority of current research. In this context, being rich in valuable compounds such as proteins, phenolics, lipids and vitamins, this study is focused in the potential activity of rapeseed press cake hydrolyzates to be used as raw materials for skin care applications. METHODS:In this study, the protein-rich press residue from the rapeseed oil industry was converted enzymatically into short-chain biologically active peptides using four protease products with varying substrate specificity -Alcalase 2.4L FG, Protex 6L, Protamex and Corolase 7089. The antioxidant, anti-wrinkle and anti-inflammatory activity of the obtained hydrolyzates were evaluated in vitro while their biocompatibility with human skin firoblasts was tested. RESULTS:All hydrolyzates were biocompatible with skin fibroblasts after 24 h of exposure, while the non-hydrolyzed extract induced cell toxicity. Alcalase 2,4L FG and Protex 6L -obtained hydrolyzates were the most promising extracts showing improved bioactivities suitable for skin anti-ageing formulations, namely anti-oxidant activity, inhibiting approximately 80% cellular reactive oxidative species, anti-inflammatory and anti-wrinkle properties, inhibiting around 36% of myeloperoxidase activity and over 83% of elastase activity. CONCLUSION:The enzymatic technology applied to the rapeseed oil industry co-stream results in the release of bioactive compounds suitable for skin care applications.
