Melanoma is one of the most serious skin cancers. The incidence of this malignant skin lesion is continuing to increase worldwide. Melanoma is resistant to chemotherapeutic drugs and highly metastatic. Surgical resection can only be used to treat melanoma in the early stages, while chemotherapy is limited due to melanoma multi-drug resistance. The overexpression of glutathione S-transferase (GST) may have a critical role in this resistance. Caffeic acid phenethyl ester (CAPE) is a natural phenolic compound, which occurs in many plants. Previous studies demonstrated that CAPE suppresses the growth of melanoma cells and induces reactive oxygen species generation. It is also known that bioactivation of CAPE to its corresponding quinone metabolite by tyrosinase would lead to GST inhibition and selective melanoma cell death. We investigated the biochemical toxicity of CAPE in combination with microsecond electropermeabilization in two human melanoma cell lines. Our results indicate that electroporation of melanoma cells in the presence of CAPE induced high oxidative stress, which correlates with high cytotoxicity. Moreover, it can disrupt the metabolism of cancer cells by inducing apoptotic cell death. Electroporation of melanoma cells may be an efficient CAPE delivery system, enabling the application of this compound, while reducing its dose and exposure time.

Introduction The central nervous system (CNS) is the most metabolically active organ characterized by high oxygen demand and relatively low anti-oxidative activity, which makes neurons and glia highly susceptible to damage by reactive oxygen and nitrogen byproducts as well as neurodegeneration. Free radicals are associated with secondary injuries that occur after a primary brain injury. Some of these free radical products include F2-Isoprostane (F2-IsoPs), malondialdehyde (MDA), 4-hydroxy-2-nonenal (4-HNE) and acrolein. Methods In this study we measured serum F2-IsoPs levels as markers of free radical activity in 10–12 week-old male Sprague-Dawley rats weighing 200–300 g, all rats (n = 10) subjected with a head injury according to the modified marmourou model, then divided into 2 groups, one group treated with CAPE (Caffeic Acid Phenethyl Ester) (n = 5) and the other not treated with CAPE (n = 5), serum levels in the two groups were compared starting from day-0 (before brain injury), day-4 and day-7. Results We found lower F2-IsoPs levels in the group that received the CAPE treatment compared to the group that did not receive the CAPE treatment. Conclusion CAPE is capable of significantly reducing oxidative stress in brain injury.

Background: This study aimed to investigate the antioxidative and anti-inflammatory effects of caffeic acid phenethyl ester (CAPE) on damage caused to cerebellum tissue by diffuse traumatic head trauma via biochemical, histopathologic, and immuno-histochemical methods. Materials and methods: Male Sprague-Dawley (300-350 g) rats were subjected to traumatic brain injury with a weight-drop device (300 g/1 m weight-height impact). Twenty-four adult rats were randomly divided into three equal groups of 8, including a control group, traumatic brain injury (TBI) group, and TBI + CAPE treatment group (10 μmoL/kg/i.p.). Cerebellum tissue samples taken from anterior lobe from all rats were taken 7 days after traumatic injury and were subjected to biochemical and histopathological analysis, as well as immunohistochemical analysis for platelet endothelial cell adhesion molecule-1 (PECAM-1) and phosphate 38-mitogen-activated protein kinase (p38 MAPK). Results: In the TBI group, the granular layer had dilatation and haemorrhage in the capillary vessels and inflammatory cell infiltration around the periphery of the blood vessels. In the TBI + CAPE group, the small capillaries in the white matter were slightly dilated, there were no inflammatory cells, and dense chromatin/ granular cells were observed in the granular layer. Also in the TBI + CAPE group, the Purkinje cells of the ganglion cell layer had ovoid nuclei, were chromatin-rich, and their extensions protruded to the molecular layer. CAPE is thought to regulate inflammation, cell damage, and angiogenetic development by affecting the PECAM-1 and p38 MAPK proteins. Conclusions: These proteins are key modulators of endothelial integrity and neuroinflammation in vessels in response to endothelial damage as well as of the proinflammatory response in the cerebellum in response to traumatic damage.
