Objectives : Determine the antidepressant activity of some fractions of the leaves of Ocimum basilicum L Methods : The experimental methods are using Force Swimming Test and Tail Suspension Test. The experiment was divided into seven groups. Negative control group (CMC 1 %), Positive control group (fluoxetine 0,052 mg/20 g BW of mice), test I group (fraction of n-hexane 50 mg/Kg BW), test II group (fraction of ethyl acetate 50 mg/Kg BW), test III group (fraction of ethanol 50 mg/Kg BW), test IV group (combination of cyproheptadine 0,0208 mg/20 g BW of mice and fluoxetine 0,052 mg/20 g BW of mice) and test V group (combination of cyproheptadine 0,0208 mg/20 g BW of mice and fraction of ethyl acetate 50 mg/Kg BW). Result : The Force Swimming Test and Tail Suspension Test showed the same result that ethyl acetate fraction was given significant difference (p<0,05) in immobilization time reduction compared to negative control and has a better immobility reduction time than other groups. Added on cyproheptadine to fluoxetine and ethyl acetate fraction groups showed an increased in immobilization time compared to fluoxetine and ethyl acetate fraction alone. Conclusion : Ethyl acetate fraction showed better antidepressant activity than another fraction through the role of serotonin.

Aim Trigonelline is a potent phytochemical present in fenugreek, which has strong anti-oxidant and phytoestrogenic activities. This study was carried out to investigate this estrogenic activity as a possible mechanisms involved in preventing the symptoms of osteoporosis in dexamethasone induced osteoporosis in rats. Materials and Methods Wistar rats were randomly divided into eight groups, six animals in each group. Osteoporosis was induced using dexamethasone 0.1mg/kg subcutaneously in rats for three times per week for 8 consecutive weeks and treatment with drugs up to 12 weeks as per the treatment schedule described. After 12 weeks, rats were sacrificed; blood samples were collected from each rat and the clear, non hemolysed supernatant sera was used for biochemical examinations. Femurs were used for Bone Mineral Density (BMD), microcomputed tomography (Micro CT), histology and biochemical examinations. Results BMD, bone micro structure, serum calcium, phosphorus level and serum estradiol levels were decreased while serum PTH levels, SAP and acid phosphatase (ACP) were elevated in dexamethasone treated rats as compared to control (p<0.01). Dexmethasone treated animals showed loss of marrow at multifocal area, cartilage and trabeculae and thinning of trabeculae (bone resorption), zone of cartilage was poorly seen and fat cells in marrow. Trigonelline showed significant improvement and prevent the progression of osteoporosis by enhancing the BMD, restoring bone physiology. Conclusion Our results confirm the estrogenic activity of triogonelline, which is responsible for its effects; still, it needs further evaluation in other animal models to provide a more conclusive view for its therapeutic usefulness in osteoporosis.

The effects and mechanisms of soybean isoflavone on osteoblast (OB) proliferation in vitro were investigated. Fifty female Wistar rats were randomly divided into five groups with 10 rats in each group. Rat OBs were separated and cultured. The first generation of OBs cultured for 48 hr at various concentrations of isoflavone were set as the experimental groups, the OBs exposed to estradiol (E 2 ) culture were considered as positive control group. The biological characterization of OBs was investigated by phase contrast microscopy and alkaline phosphatase (ALP) histochemistry. The concentrations of interleukin (IL-1), osteoprotegerin (OPG), transforming growth factor (TGF), granulocytemacrophage colony-stimulating factor (GM-CSF), and vascular endothelial growth factor (VEGF) in isoflavone culture solutions were determined. Proliferation rate of OBs was increased in experimental group comparing that in the blank group. ALP activity in experimental group was higher than that in blank group. No significant differences of ALP activity were observed between E 2 culture group and isoflavone group at concentrations of 10 −5 and 10 −7 mM (P > 0.05). Furthermore, in the experimental groups at low isoflavone concentrations, the concentrations of OPG, TGF, and VEGF were increased and positively correlated with OB proliferation. However, the concentrations of IL-1, GM-CSF were decreased at higher concentration of isoflavone and were negatively correlated with OB proliferation. Soybean isoflavone could promote the growth and proliferation of rat OB, it might act as the stimulator of OPG, TGF, and VEGF pathway, and the inhibitor of IL-1, GM-CSF pathway as well.
