Fluoroquinolones (FQs) have been identified recently as potent inhibitors of pancreatic lipase (PL). The aim of this study was to synthesize novel FQs and triazolofluoroquinolones (TFQs) and to evaluate them in vitro with respect to their antilipolytic efficacy and potency properties. The PL-IC values of 12 FQs and TFQs (3 (a-c)-6 (a-c)) were in the range of 12.5-189.1 μm. These values are further supported by docking studies. The suggested association between obesity and colorectal cancer initiated the evaluation of antiproliferative activity of the new FQs and TFQs against a panel of obesity-related colorectal cells (HT29, HCT116, SW620 CACO2, and SW480). Sulforodamine B colorimetric assay revealed that some derivatives exhibited unselective cytotoxicity against HT29, HCT116, SW620 CACO2, and SW480. Remarkably, FQ 4a's selective cytotoxicity against HCT116 was found valuable with IC value of 4.2 μm which exceeds that of cisplatin with a substantial selective cytotoxicity in periodontal ligament fibroblasts. In conclusion, FQ and TFQ derivatives may unveil new antiobesity and anticancer agents in the future.

Introduction: Evidence suggests that hypoxia and high-intensity exercise training can increase apoptosis of lung cells and Adiantum capillus-veneris (Ac-v) extract can have anti-apoptotic effects. Thus, the aim of the present study was to investigate the effect of chronic hypoxia and the (Ac-v) extraction as a supplement on TNF-a and P53 protein expression as well as the respiratory surface. Material and methods: 24 healthy Wistar rats (age = 4 weeks, weight = 72 = 9 gr) were trained using interval training for 6 weeks followed by a 3-week stay in hypoxia conditions. Half of the hypoxia samples received 500 ml/gr/per body weight daily (Ac-v) within 3 weeks of hypoxia. At the end, the lung tissue was removed for histological and immunohistological analysis. Results: After 3 weeks of hypoxia exposure following 6 weeks of exercise, expression of P53 and TNF-a increased and the respiratory surface decreased (p ≤ 0.05). After 3 weeks of taking the Ac-v extract during hypoxia exposure, reduced P53 and TNF-a expression and the increased respiratory surface were observed (p ≤ 0.05). Conclusions: Chronic hypoxia may be considered as a strong stimulus leading to the expression of proteins involved in apoptosis and tissue disruption. However, our findings suggest that the antioxidative properties of Ac-v extract could decrease the destructive structural and molecular events that happen along with hypoxia exposure or intense exercise training.

Objective: To investigate the effect of hydro-ethanolic fraction of leaves of M. longifolia (J. Koenig) on high fat diet induced metabolic disorder. Methods: The M. longifolia extract was obtained from the successive extraction followed by fractionation. Ex vivo glucose uptake study was performed. Body weight and blood glucose was checked at regular intervals. After euthanasia serum biochemical parameters, lipid parameters were estimated. Organs were isolated, weighed and fixed in formalin for histopathological analysis. Results: Compared to control, M. longifolia leaf fractions showed significant glucose uptake inhibition in ex vivo studies (rat hemi diaphragm and rat intestinal glucose uptake assay). HFD group showed significant weight increase as compared to normal control. Blood glucose level and glycosylated haemoglobin was found elevated in disease group (obesity and diabetic group). Significant increase in lipid profile of disease group aggravates the diabetic condition. Animal treated with M. longifolia leaf fractions (400mg/kg) helps in normalisation of liver and white adipose tissue. Conclusion: Metabolic syndrome is an agglomeration of disorders inclusive of increased blood glucose, excess body fat, anomalous triglyceride and cholesterol level, which on joint occurrence leads to diabetes and increased risk of heart disease. Our data suggest that treatment with M. longifolia to HFD + STZ group alleviates the diabetes and helps in control of obesity. It may be due to presence of various bioactive constituents.
