Phytochemicals might offer economic therapies, which are an alternative especially for low-income developing countries. Anemone coronaria has been used extensively as antineuralgic and antirheumatic in the folk medicine of Lebanon. Thus, the aim of this study was to investigate the antinociceptive potential of A. coronaria and its active principles against diabetic-neuropathy pain in a mouse-model. The study also aimed to achieve a bio-guided fractionation and to isolate the most active principles in A. coronaria herb. Bio-guided fractionation used reversed phase-HPLC, and 1H and 13C NMR for the identification of the most active fraction of A. coronaria, which was rich in alkaloids. This isolated alkaloid rich fraction contained catalin (21.1%), thaliporphine (14.1%), and glaucine (compound 3) (63.4%). A. coronaria extract (50, 100, and 200 mg/kg), the isolated alkaloid rich fraction (70 mg/kg), and glaucine (25, 50, and 100 mg/kg) showed significant reduction in acute and subchronic hyperglycemia, with significant increase in glutathione and catalase levels, along with normalization of HbA1c and LPO levels (n=7 per group, p≤0.05). A. coronaria extract (50, 100, and 200 mg/kg), the isolated alkaloid rich fraction (70 mg/kg), and glaucine (25, 50, and 100 mg/kg) also showed significant antinociceptive amelioration in thermal hyperalgesia pain latencies utilizing hot plate and tail flick tests. The highest doses of A. coronaria extract (200 mg/kg), isolated alkaloid rich fraction (70 mg/kg), and glaucine (100 mg/kg) showed a significant elevation in mechanical allodynia pain thresholds using Von-Frey-filaments. The results revealed that A. coronaria herb along with its most active alkaloid rich fraction and constituent (glaucine) possessed significant antihyperglycemic and antinociceptive potentials. These findings may lead to a future use of A. coronaria in the management of diabetic-neuropathy pain.

Acmella oleracea is a herbaceous plant native to the Amazon region that is currently arousing industrial interest due to its potential gastronomic, cosmetic and pharmaceutical uses. Accordingly, the objective of the current study was to test the influence of temperatures on A. oleracea seed germination and seedling vigor. A completely randomized design (CRD) with five treatments and five replicates was used. Treatments consisted of temperatures: 23, 26, 29, 32 and 35 ºC. The study was conducted in BODtype climate chambers with relative humidity above 90% and a 12 h photoperiod. Germination, germination speed index (GSI), average germination time, root length and number of normal and abnormal seedlings were recorded. Temperature influenced all analyzed variables, except average germination time and number of normal/abnormal seedlings. A quadratic model best described the relationship between dependent and independent variables. In general, maximum germination values, and GSI and root lengths were obtained at temperatures close to 29 °C, while the lowest observed values for these variables occurred at extreme temperatures (23 and 35 °C). The highest percentage of germinated seeds occurred on the second day. Temperatures above 29 ºC induced a significant reduction in the A. oleracea seedling root lengths.

The present study analyzed the effects of different concentrations of Acmella oleracea crude ethanolic extract (EEAO) on the development of germ cells from semi-engorged Amblyomma cajennense females in order to evaluate the potential of this natural chemical as a strategy to control these important ectoparasites. A hundred semi-engorged females were divided into five groups (duplicates) (10 animals/group): Control 1 (distilled water); Control 2 (solvent ethanol 50% and DMSO 1%); and Treatment I to III (3.1, 6.2, and 12.5 mg/mL of EEAO, respectively).For the exposure of the ticks to the extract was used the Adult Immersion Test. After the exposition, the ovaries were removed and submitted to histological analysis using Harris hematoxylin and aqueous eosin. The histochemical tests were performed using PAS and Bromophenol blue staining techniques, for the detection of total polysaccharides and total protein, respectively.The extract caused significant alterations in the oocytes, including changes in the shape of the cells, disorganization, and cytoplasmic vacuolation, decrease in the number of yolk granules and germ vesicle fragmentation. These alterations were more intense in the oocytes in initial developmental stages (I and II). The results obtained in this study confirm the cytotoxic potential of the ethanolic extract of A. oleracea on the germ cells of A. cajennense females, opening up the possibility to use this extract as an alternative to control these ectoparasites.
