Medicinal plants have been recognized as hosts of high diverse endophytic microorganisms, including fungi that produce secondary metabolites with biological activity. Two biomes in Brazil, Pantanal (wetland), and Cerrado (savannah), are known as biodiversity hotspots, and despite their importance as a reservoir for several species, knowledge about the fungal biodiversity in these biomes is very limited. Fungal endophytic communities associated with leaves and petioles of the medicinal plants Vochysia divergens (from Pantanal) and Stryphnodendron adstringens (from Cerrado) were analyzed and studied for their antimicrobial activity against human and plant pathogens. A total of 1,146 isolates of endophytic fungi were obtained from plants collected in January and June of 2016 and grouped into 124 morphotypes. One isolate of each morphotype was identified by sequencing of internal transcribed spacer (ITS) region of the rDNA gene, which revealed the presence of 24 genera, including 3 possible new genera, and 48 taxa. Differences in the endophytic community according to the biomes were observed concerning the analyzed morphotypes. However, when we analyzed the diversity of genera and richness, they were similar for both plants, with Diaporthe, Phyllosticta, and Neofusicoccum as dominant genera. In addition, the community composition of V. divergens differs according to the analyzed plant tissues (petiole and leaf). These data suggested that both, the plant species and plant tissues play a role in the composition of endophytic community. As regards the biotechnological potential, 5 isolates showed activity against the phytopathogens Phyllosticta citricarpa, Colletotrichum abscissum, and Fusarium verticilioides, and 8 isolates showed high activity against clinical pathogens and were selected for the production of crude extract in different culture media. Extract from cultivation of Diaporthe sp. LGMF1548 and LGMF1583 and Neofusicoccum brasiliense LGMF1535 showed activity against methicillin-resistant Staphylococcus aureus, Klebssiella pneumonia, and Candida albicans. In addition, extracts of Diaporthe cf. heveae LGMF1631 inhibited 90% of the mycelial growth of the P. citricarpa and 70% of C. abscissum and may represent an alternative to be used in the biological control of these phytopathogens. Future research will focus on the chemical characterization and structural elucidation of these bioactive compounds.

Colletotrichum, Aspergillus, Paecilomices, Xylaria, Phomopsis e Pestalotiopsis, em meio de cultura batata-dextrose, acrescido de 0,2% de extrato de levedura, a 24±1° C por 21 a 28 dias. O micélio foi separado por filtração a vácuo e os constituintes fixos extracelulares (meio de cultura fermentado) e intracelulares (micélio) foram extraídos com metanol sob refrigeração a 4 o C. Os ensaios para determinação da atividade antimicrobiana foram realizados em placas de ELISA de 96 poços. As amostras foram divididas em 6 grupos: GC-cimento de agregado trióxido mineral branco (MTA) Angelus ® e G2 a G6-parte do pó de MTA foi substituído, em diferentes concentrações (5%,10%,15%, 20%, 25%), pela fração extraída do cultivo do fungo endofítico P. microspora. O gênero Pestalotiopsis mostrou-se mais efetivo nos testes antimicrobianos, especificamente o P. microspora, com a concentração inibitória mínima de 0,125 para E. feacalis, 0,25 para S. aureus, P. aeruginosa, C. albicans e 0,5 para C. tropicalis. Na avaliação da atividade antimicrobiana do cimento experimental, houve atividade para o S. aureus, E. coli, P. aeruginosa e E. feacalis nos grupos GC, G5 (MTA + 20%) e G6 (MTA + 25%). O G6 apresentou os maiores valores e não houve diferença significativa entre os grupos de E. coli, S. aureus e E. faecalis. A adição do extrato de polissacarídeos intracelulares do P. microspora melhorou a atividade antimicrobiana do MTA, constituindo-se em uma alternativa promissora para o uso odontológico.

Aims: The present study was carried out to determine the diversity of endophytic fungi that colonize the leaves of Psidium guajava, and to evaluate their antagonistic activity against Fusarium oxysporum f.sp. cubense and Mycosphaerella fijiensis which are the two main phytopathogens of banana plants. Place and Duration of Study: The research was carried out at Microbiology Laboratory, Faculty of Sciences, University of Yaoundé I and Microbiology Laboratory, Faculty of Biotechnologies, University of Agronomic Sciences and Veterinary Medicine Bucharest, between April 2018 and February 2020. Methodology: Fragments of surface sterilized leaves of Psidium guajava were inoculated on Potato Dextrose Agar supplemented with chloramphenicol. The isolated and purified endophytic fungi were identified based on their macroscopic and microscopic characters using a mycological atlas as guide. The non-sporulating isolates were identified by comparing the ITS regions of their DNA to those of known fungi registered in the GenBank database. The antagonistic activity of the endophytic fungi isolated against Fusarium oxysporum and Mycosphaerella fijiensis was screened using dual culture method. Results: A total of 28 endophytic fungal were isolated from the leaves of Psidium guajava corresponding to a colonization frequency of 33.33%. These isolates were identified as: Aspergillus sp., Botryosphaeria sp., Fusarium sp., Neoscytalidium sp., Xylaria sp., Phyllosticta capitalensis, Cercospora apii, Xylaria longipes, Phomopsis sp., Phomopsis asparagi, Aspergillus versicolor, Pallidocercospora thailandica, and Xylaria grammica that belonged to the Deuteromycota and Ascomycota divisions. These endophytic fungi inhibited the growth of Fusarium oxysporum f.sp. cubense and Mycosphaerella fijiensis with the percentage inhibition varying respectively from 23.25% to 73.52% and from 21.36% to 100%. The species Botryosphaeria sp., Phomopsis sp., Phomopsis asparagi, and Xylaria longipes exhibited the greatest activity. Conclusion: The leaves of Psidium guajava have a fairly varied diversity of endophytic fungi. These endophytic fungi can serve as potential biological control agents against Panama and Sigatoka diseases of banana and also would produce secondary metabolites with antifungal properties.
