Antidiarrhoeal and antispasmodic activity of leaves of Syzygium cumini L. (Myrtaceae) mediated through calcium channel blockage

Abstract: Syzygium cumini L. Skeels (Myrtaceae) commonly known as jambolan is used as traditional medicine to treat gastrointestinal disorders in children in Brazil. This work is one of the first to evaluate the antidiarrhoeal and antispasmodic activity of the standardized extract of S. cumini leaves (HESc) in experimental models in vitro and in vivo rodents. Mice pre-treated with HESc (100, 250 and 1000 mg/kg) and atropine (1.0 mg/kg) had reduced intestinal transit velocity of 11.0; 23.2 and19.1%, respectively compared… Show more

“…For this, verapamil (1 µM) was employed as a positive control. This chemical has been identified as a calcium channel blocker for L-type dependent VGC channels [35]. Our results indicate that EoWs prevented calcium entry through the VGC channels and, as a result, calcium release from the endoplasmic reticulum.…”

“…The results indicate that AEJO (0.1, 0.3, and 1 mg/ml) added in a cumulative fashion diminished the maximum tone induced by low K + , while demonstrating a weak inhibitory effect on high K + in a dose-dependent manner, thus indicating that the inhibitory effect probably passed through K + channel opening activity (15). All doses of AEJO completely inhibited the maximum response to CaCl 2 , similar to the effect of verapamil, a positive control known for blocking voltage-dependent L-type channels (19). Like non-competitive antagonists, AEJO blocks Ca 2+ channel pathways.…”

“…EH-FAB concentrations were used in multiples of three, with the maximum concentration being 729 µg/mL since it is the maximum concentration used in experiments with isolated organs. When the observed effect was larger than 50% at 729 µg/mL, it was sought the concentration that would provide a 0% effect (Monteiro et al, 2018(Monteiro et al, , 2020.…”
