Chemical investigation of the aerial parts of Ammania aegyptiaca ethanol extract (AEEE) showed high concentrations of polyphenol and flavonoid content, with notable antioxidant activity. Undescribed acylated diglucoside flavonol myricetin 3-O-β-4C1-(6”-O-galloyl glucopyranoside) 7-O-β-4C1-glucopyranoside (MGGG) was isolated from the aerial parts of AEEE, along with four known polyphenols that had not been characterized previously from AEEE. The inhibitory effects of MGGG, AEEE, and all compounds against α-amylase, pancreatic lipase and β-glucosidase were assessed. In addition, molecular docking was used to determine the inhibition of digestive enzymes, and this confirmed that the MGGG interacted strongly with the active site residues of these enzymes, with the highest binding free energy against α-amylase (−8.99 kcal/mol), as compared to the commercial drug acarbose (−5.04 kcal/mol), thus justifying its use in the potential management of diabetes. In streptozotocin (STZ)-induced diabetic rats, AEEE significantly decreased high serum glucose, α-amylase activity and serum liver and kidney function markers, as well as increasing insulin blood level. Moreover, AEEE improved the lipid profile of diabetic animals, increased superoxide dismutase (SOD) activity, and inhibited lipid peroxidation. Histopathological studies proved the decrease in pancreas damage and supported the biochemical findings. These results provide evidence that AEEE and MGGG possess potent antidiabetic activity, which warrants additional investigation.

New acylated diglucoside flavonol myricetin 3-O-β-4C1-(6"-O-galloyl glucopyranoside) 7-O-β-4C1-glucopyranoside (MGGG) was isolated from aerial parts of AEEE along with four additional known phenolics, not characterized previously from AEEE. Moreover, powerful inhibitory effects of MGGG, AEEE, and all isolates against α-amylase, pancreatic lipase and β-glucosidase, were assessed. In addition, flexible molecular docking was used to reveal the inhibition towards digestive enzymes and confirmed that the MGGG interacted strongly with the active site residues of these enzymes with the highest binding free energy against β-glucosidase (DG=-8.98 kcal/mol) compared to the commercial drug Acarbose, thus justifying its dual management of diabetes and obesity. In streptozotocin (STZ) induced diabetic rats, AEEE significantly decreased high serum glucose, α-amylase activity, liver and kidney function markers and increased insulin level. Moreover, it improved lipid profile due to diabetes with increased SOD activity and inhibited of TBARS formation. Consequently, AEEE and MGGG are found useful in controlling the secondary complications associated with type 2 diabetes mellitus. Histopathological studies proved the decrease in the pancreas damage and agreed with the biochemical findings. These results provide evidence that AEEE and MGGG have potent antidiabetic activity, which warrants additional investigations.

Vascular dementia (VaD) is a major factor for the progress of stroke and other memory disorders. It is the second leading cause of death. The burden of VaD is higher in the aged population (>65 years). The progression of VaD occurs with lifestyle modifications i.e., fast food, smoking, and alcohol. The medicines for the treatment of VaD are limited. The palm oil is one of the rich sources of beta-carotene (BC). The present research designed to investigate the potential role of palm oil mill effluent derived BC in experimental model of diabetic VaD. The diabetic VaD was induced by administration of nicotinamide (NA, 50 mg/kg; i.p.) followed by streptozotocin (STZ, 50 mg/kg; i.p.). The test compound i.e., BC (50 and 100 mg/kg) and reference compound donepezil (1 mg/kg) were administered orally for 15 consecutive days. The changes of cognitive patterns i.e., escape latency time (ELT) and time spent in target quadrant (TSTQ) was assessed by Morris water maze (MWM) test. Besides the changes of neurotransmitter i.e., acetylcholinesterase (AChE) was estimated in brain (hippocampus, cerebellum, entorhinal cortex, amygdala and septum) samples. The administration of STZ caused the significant changes of cognitive functions (increased ELT and decreased TSTQ) as indicated in the development of VaD when compared to normal group. The treatment of BC was ameliorated the cognitive dysfunctions against the STZ associated cholinergic neurotransmitter (elevated AChE) changes. The effects were similar to donepezil treatment group. Hence, it proved that BC possesses the potential therapeutic effects in the management of diabetic VaD due to its potential anti-cholinergic effects.
