Anticonvulsant Effects of Fuzi Total Alkaloid on Pentylenetetrazole-Induced Seizure in Mice

Abstract: Abstract. We investigated the anticonvulsant effect of acute Fuzi total alkaloid (FTA) in seizure induced by the GABA A -receptor antagonist pentylenetetrazole (PTZ). FTA significantly increased the seizure latency and decreased the mortality in PTZ-treated mice. Administration of PTZ increased c-Fos expression in the hippocampus, medial prefrontal cortex, and piriform cortex; and this PTZ-induced effect was inhibited by FTA in a dose-dependent manner. Furthermore, the effects of FTA on PTZ-induced seizure and… Show more

“…However, Hippocampus is most widely analyzed as a representative brain region in PTZ-induced seizure mice [11], [17], [25][27]. In our immunohistochemical study c-Fos expression was also significantly increased by PTZ in the dentate gyrus, CA1, CA3 and CA4 of the hippocampus.…”

“…Animals were killed 2 h after injection of PTZ, the brains were removed rapidly, flash frozen in isopentane and stored at −80°C. Immunohistochemistry for c-Fos protein was carried out as described by previous reports from our group and others [17], [20]. Serial coronal sections from forebrain to brainstem, 30 μm thick, were cut in a cryostat.…”

“…Epilepsy is a condition that affects the brain and is caused by an abnormally large discharge of neurons, which are sudden, recurrent, and transient . FTA notably prolongs the latency time of epileptic seizures and decreases the mortality of pentetrazole‐treated groups . Ameri's findings suggest that aconitine acts as an anticonvulsive.…”
