In this study, we synthesized the cobalt(II) chelation free complexes (1–3) with the general formula of [CoCl2L2] containing monodentate ligands (L=2‐methylbenzimidazole (L1), 2‐methylbenzoxazole (L2), and 2‐methylbenzothiazole (L3) and fully characterized by UV‐vis, FT‐IR spectroscopy, and the crystal structures of 1–3 were determined by X‐ray crystallography. Complexes 1–3 showed substantial antibacterial activity against tested bacteria. Complexes 2 and 3 were more active against S. aureus with the Minimum Inhibitory concentration (MIC) value of 12.5 μg/mL compared to other bacteria. Complexes 1–3 exhibited strong synergy with ampicillin (AMP) against Methicillin Resistant S. aureus strains (MRSA). Interestingly, complexes 2 and 3 in combination with AMP showed the membrane permeabilization of MRSA. This indicates that complexes 2 and 3 in combination with AMP had bacterial membrane disruption as a possible mechanism of action.

In searching for new therapeutic agents for treating American trypanosomiasis and Human African trypanosomiasis, four nitroheterocyclic acylhydrazones of general formulae [R1‐CH=N‐NH‐C(O)‐(5‐C4H2X)] (where R1 = ferrocenyl or cyrhetrenyl, and X = O or S) have been synthesized and characterized by spectroscopic techniques. Comparative studies of their stability by 1H‐NMR and UV–Vis experiments were reported. Single‐crystal X‐ray diffraction confirmed the molecular structures of NF‐1 and NT‐2. Their X‐ray crystal structures reveal that both adopt an E‐configuration on the C=N moiety. Regarding the ‐NH‐C(O)‐ bond, the structure of NF‐1 confirmed a trans conformation, while NT‐2 exhibited a cis‐amide conformation. The cyclic voltammetry and electron spin resonance (ESR) experiments were conducted to study the electrochemical behavior of N‐acylhydrazones. The antiparasitic activities of compounds against Trypanosoma cruzi (epimastigotes) and Trypanosoma brucei (trypomastigotes) revealed that cyrhetrenyl complexes were more effective than their ferrocenyl analogs. The cyrhetrenyl derivative NT‐2 (EC50 = 2.25 μM) showed activity against T. brucei comparable to the standard drug nifurtimox (Nfx, EC50 = 3.56 μM). The ferrocenyl compound NT‐1 (>200 μM) was at least two times less cytotoxic than the Nfx (88.7 μM) against the L6 rat skeletal myoblast cell line and exhibited a selectivity like Nfx toward T. brucei. Density functional theory (DFT) calculations were utilized as an approximation to explain the impact of organometallic and heterocyclic rings on antiparasitic activities. This study supported the experimental results, confirming that the cyrhetrenyl fragment in N‐acylhydrazone derivatives plays a significant role in the antitrypanosomal activity, which can be attributed to an increase in positive charge on the metal.

The phosphine ligand (Ph2PCH2N(CH3)(CH2)2Ph, PNMPEA) obtained by the reaction of the (hydroxymethyl)diphenylphosphine with naturally occurring alkaloid N‐methylphenethylamine, was used to synthesize the half‐sandwich iridium(III) (Ir(η5‐Cp*)Cl2Ph2PCH2N(CH3)(CH2)2Ph, IrPNMPEA) and ruthenium(II) (Ru(η6‐p‐cymene)Cl2Ph2PCH2N(CH3)(CH2)2Ph, RuPNMPEA) complexes. They were characterized using a vast array of methods, including 1D and 2D NMR, ESI(+)MS spectrometry, elemental analysis, cyclic voltammetry (CV), electron spectroscopy in the UV‐Vis range (absorption, fluorescence) and density functional theory (DFT). The initial antimicrobial activity in vitro toward Gram‐positive and Gram‐negative bacterial strains was examined, indicating that both complexes are selective towards Gram‐positive bacteria, e.g., Staphylococcus aureus, where the IrPNMPEA has been more bactericidal compared to RuPNMPEA. Additionally, the interactions of these compounds with various biomolecules, such as DNA (ctDNA, plasmid DNA, 9‐ethylguanine (9‐EtG), and 9‐methyladenine (9‐MeA)), nicotinamide adenine dinucleotide (NADH), glutathione (GSH), and ascorbic acid (Asc) were described. The results showed that both Ir(III) and Ru(II) complexes accelerate the oxidation process of NADH, GSH and Asc that appeared to occur by an electron transfer mechanism. Interestingly, only IrPNMPEA leads to the formation of various biomolecule adducts, which can explain its higher activity. Furthermore, RuPNMPEA and IrPNMPEA have been interacting with the DNA through weak noncovalent interactions.
