Antibody to Hepatitis B Virus Induced by Injecting Antibodies to the Idiotype

Abstract: Anti-idiotype reagents that recognize a common idiotype associated with antibody to hepatitis B surface antigen (anti-HBs) were used to induce anti-HBs in mice. The anti-idiotype-induced anti-HBs was found to recognize the group-specific a determinant of hepatitis B surface antigen and to express an interspecies idiotype. These findings suggest that anti-idiotypes may be useful as vaccines or vaccine primers.

“…(11) found a monoclonal antibody specific for A48 Id borne by ABAC48 or [32-6 fructosan binding protein able to elicit an anti-[32-6 fructosan response in BALB/c mice. Strong antiviral antibody responses were also obtained in mice by injection of rabbit anti-Id antibodies against monoclonal antibodies specific for HBV (12) or rabies (13).…”

“…Jerne et al (20) classified anti-Id Abs in two categories: Ab^a, or conventional anti-Id Abs and Abpß, anti-Id Abs carrying the internal image or the antigens. There are numerous 132 studies indicating that anti-Id Abs enhance the immune responses against bacterial (8), viral (22)(23)(24)(25)(26) and parasitic antigens (27). However, there are no clear cut immunochemical or structural criteria to distinguish Abpoc from Ab2ß (24).…”

“…Anti-idiotypic antibodies have been advocated as alternatives to traditional vaccines (Fields & Greene, 1982;Reagan et al, 1983;Ertl & Finberg, 1984;Hay et at., 1984;Kennedy et aL, 1984b;GelI & Moss, 1985 ;Utydehaag & Osterhaus, 1985) but in viral systems have been so far without notable success. In our own investigations with anti-idiotypic reagents against monoclonal antibodies to HSV-I glycoproteins, we have observed only weak cellular responses and no measurable antibody following administration to mice in vivo (Lathey et al, 1986(Lathey et al, , 1987.…”
